A helmintosporiose causada por Exserohilum turcicum é uma das doenças mais sérias que ocorrem em folhas de plantas de milho (Zea mays L.). O progresso desta doença, característica de clima tropical, ocorre principalmente pelo aumento no tamanho das lesões e não em seu número. Essas lesões, com características variáveis, que dependem do genótipo da interação patógeno-hospeiro, representam os sítios de infecção. Dessa forma, o trabalho realizado objetivou caracterizar esses sítios de infecção com base em mecanismos estruturais e bioquímicos de defesa expressos pela planta, bem como verificar diferenças entre as respostas de defesa em materiais resistente e suscetível ao patógeno. Plantas das linhagens de milho F64A e F352, suscetível e resistente a E. turcicum foram cultivadas em câmara-de-crescimento com fotoperíodo de 14 h de luz e temperatura de 25°C. Após 14 dias, a 4ª folha verdadeira de cada planta foi inoculada com uma suspensão de 1 x104 esporos/ml do patógeno crescido em meio sólido do tipo LCH por 14 dias no escuro e à 25°C. As amostragens foram realizadas a diferentes intervalos de tempo durante as primeiras 48 h após a inoculação, para observação do desenvolvimento do fungo nos tecidos e da ocorrência de lignificação. Amostras também foram obtidas 12 dias após a inoculação, para verificar: o conteúdo de compostos fenólicos, as atividades de β-1,3 glucanases, quitinases e fenilalanina amônia-liase, as atividades e o padrão isoenzimático de peroxidases, a presença de compostos fungitóxicos in vitro e a quantificação do fungo nos tecidos através de dosagem de ergosterol. Os resultados obtidos mostraram que no material resistente, o sítio de infecção (denominado de sítio de infecção primário) é representado por sintomas do tipo pontos cloróticos resultantes da penetração do patógeno nos vasos do xilema e restrita colonização nos tecidos do mesófilo ao redor, verificando-se a ocorrência de lignificação, acúmulo de compostos fenólicos e atividade acentuada de β-1,3 glucanases e quitinases. No material suscetível há dois tipos de sítios de infecção. No primeiro (sítio de infecção primário), representado por sintomas do tipo pontos cloróticos, o patógeno penetra os vasos do xilema e há moderada colonização dos tecidos do mesófilo ao redor, verificando-se atividades moderadas de β-1,3 glucanases e quitinases. Após a formação do sítio de infecção primário, o patógeno cresce pelo interior dos vasos do xilema e coloniza intensamente os tecidos do mesófilo distantes do ponto de penetração, resultando no sítio de infecção secundário e na formação de lesões necróticas, onde se verifica o acúmulo de compostos fenólicos e atividades moderadas de β-1,3 glucanases e quitinases.

Characterization of infection sites of Exserohilum turcicum (Pass.) Leonard & Suggs in different maize genotypes (Zea mays L.), based upon plant defense mechanisms

Northen leaf blight caused by Exserohilum turcicum is one of the most important diseases in maize plants. The progress of this tropical disease occurs mainly by increases in lesion size instead of numbers. Lesions, which exhibit variable characteristics depending upon the host-parasite interaction, represent the infection sites. Thus, the present work was carried out to characterize these infection sites on maize leaves based on structural and biochemical defense mechanisms as well as to study these mechanisms in resistant and susceptible plants to E. turcicum. The maize inbreds F64A e F352, susceptible and resistant to E. turcicum, respectively, were grown in growth chambers under a photoperiod of 14 h of light at 25°C. After 14 days, the 4th leaf of each plant was inoculated with a conidium suspension (1x104 spores/ml) of the pathogen grown on LCH media. Leaf tissue was sampled at different time intervals during the first 48 h after inoculation, in order to follow fungal development and lignification in the tissue. Samples were also obtained 12 days after inoculation to measure the amount of phenolic compounds, and activities of β-1,3 glucanase, chitinase and phenylalanine ammonia-lyase, besides the activity and electrophoretic pattern of peroxidases in the tissues. These tissue samples were also used to test for the presence of fungitoxic compounds through in vitro bioassays and to determine the amount of fungal biomass inside the leaf using ergosterol analysis. The results showed that in the resistant maize inbred, the infection site (called "primary infection site") was represented by chlorotic spots due to the penetration and growth of the fungus in the xylem vessels and restricted colonization of the mesophyll cells around it. The restriction of fungal colonization was accompanied by host tissue lignification, increases in phenolic compounds and high β-1,3 glucanase and chitinase activities. In the susceptible inbred there were two types of infection sites. The first one, the primary infection site, was represented by chlorotic spots, due to pathogen penetration and colonization of the xylem vessels, resulting in moderate β-1,3 glucanase and chitinase activities. After the formation of the primary infection site, the pathogen kept growing inside the xylem vessels and was able to heavly colonize the mesophyll tissue far away from the penetration area, given rise to the "secondary infection site" characterized by the production of necrotic lesions, exhibiting increases in the content of phenolic compounds and moderate activity of β-1,3 glucanase and chitinase.
