O trabalho teve como objetivo o estudo ao estímulo à embriogênese somática em linhagens de calos de laranja 'Seleta Vermelha', laranja 'Valência', laranja 'Caipira', limão ‘Cravo’ e tangerina 'Cleópatra' linhagens 1 e 2, pela utilização de diferentes fontes de carboidratos. Nas linhagens de calos, com baixo potencial embriogênico também estudou-se o efeito do inibidor de auxina 7 - azaindole (AZI), e densidade de plaqueamento de calos. O meio de cultura utilizado no estímulo à embriogênese somática, foi descrito por Murashige & Tucker (1969), substituindo-se a fonte de carboidrato por maltose, lactose, galactose, sacarose, glicose, nas concentrações de 18; 37; 75; 110 e 150 mM, ou glicerol, nas concentrações de 6; 12; 24; 36; 50 mM. Os resultados obtidos, demonstraram que a maior eficiência para o estímulo à embriogênese somática ocorreu em meio de cultura suplementado com maltose, galactose ou lactose, para as linhagens de calo de laranjas 'Valência' e 'Caipira', e tangerina 'Cleópatra' linhagem 1. Para as linhagens de calos de limão 'Cravo' e tangerina 'Cleópatra' linhagem 2, foram testados o inibidor de auxina, AZI, e a densidade de plaqueamento de calo, os quais demonstraram estar na dependência do açúcar para induzir a formação dos embriões somáticos. Pela análise histológica, pode-se verificar que embriões somáticos de limão 'Cravo' e das tangerinas 'Cleópatra' linhagem 1 e 2, apresentaram ausência de meristemas caulinar e/ou radicular, má formação da protoderme e do procâmbio. A histodiferenciação dos embriões somáticos, e a conversão em plantas foram obtidos para as laranjas 'Valência' e 'Seleta Vermelha', sendo observado um maior número de plantas nos embriões somáticos que permaneceram em meio de cultura de pré-germinação com 44 mM de sacarose. Com relação a concentração de ácido giberélico no meio de cultura para a germinação, obteve-se para a laranjas 'Valênca' e 'Seleta Vermelha', 23% e 51,4% de plantas germinadas em meio de cultura com concentração de 1,4 µM ácido giberélico, 17% e 60% de plantas germinadas e meio de cultura com concentração de 2,8 µM ácido giberélico, e 14% e 51,4% de plantas germinadas e meio de cultura com concentração de 1,4 µM ácido giberélico

This work aimed to study the induction somatic embryogenesis induction in callus lines of 'Seleta Vermelha', 'Valência' and Caipira sweet oranges, Rangpur lime and Cleopatra mandarin, lines 1 and 2, using different carbohydrate sources. In callus lines with low embryogenic potential, the effect of the auxin inhibitor 7 -azaindole (AZI) and callus platting density were studied as well. The culture medium used for induction of somatic embryogenesis was described by Murashige & Tucker (1969), exchanging the carbohydrate source by maltose, lactose, galactose, sucrose or glucose at concentrations of 18; 37; 75; 110 ou 150 mM, or glycerol, at concentrations of 6; 12; 24; 36 ou 50 mM. The results obtained showed that the highest efficiency for somatic embryogenesis stimulation occurred in the culture medium supplemented with maltose, galactose or lactose, for callus lines of 'Valencia' and Caipira sweet orange and Cleopatra mandarin, lines 1. For lines Rangpur lime and Cleopatra mandarin line 2, the auxin inhibitor AZI and the callus platting density were tested, demonstrating that the stimulation of somatic embryogenesis depends on the sugar. Histological analysis verified that somatic embryos of Rangpur lime and Cleopatra mandarin, lines1 and 2 had absence of shoot and/or root deformation of protoderm and procambium. Histodifferentiation of somatic embryos and conversion in plants were obtained for 'Seleta Vermelha' and 'Valencia' sweet oranges, with a higher number of plants from somatic embryos which were maintained in pre-germination culture medium whit 44 mM sucrose. The use of gibberellic acid in the culture medium for germination resulted in 23% and 51.4% of germinated plants for 'Valencia' and 'Seleta Vermelha' sweet oranges in culture medium with 1,4 µM gibberellic acid, and 17% and 60% of plants germinated in culture medium with 2,8 µM gibberellic acid, and 14% and 51,4% of plants germinated in culture medium with 1,4 µM gibberellic acid
