O trabalho foi desenvolvido em três etapas experimentais. A primeira consistiu da avaliação, em campo de 90 genótipos de soja, sendo 68 pertencentes ao grupo de maturação precoce e 22 ao grupo tardio, quanto à variação na capacidade de nodulação da planta hospedeira. A inoculação dos genótipos foi feita com a mistura de três estirpes de B. japonicum: SEMIA-587, SEMIA-5019 e CB-1809. Os caracteres avaliados foram número e peso de nódulos, atividade da nitrogenase (ARA) e produtividade. O tratamento estatístico dos dados incluiu análise multidimensional. Os resultados revelaram diferenças genotípicas entre os materiais testados, indicando o potencial genético da leguminosa para a fixação do N2. Não foi detectada correlação entre dados de nodulação e produtividade. A segunda etapa experimental foi conduzida em casa de vegetação com o objetivo de avaliar a resposta da bactéria dentro e entre as três estirpes de B. japonicum, inoculadas separadamente no genótipo Bossier (precoce) e IAC-9 (tardio). Os dados foram agrupados em classes de frequência da ARA e do tamanho de 60 nódulos individuais. Os resultados mostraram comportamentos diferenciais dentro das estirpes para ambos os caracteres avaliados, atribuídos à variação genética intrínseca da estirpe. A CB-1809 apresentou curva de distribuição normal, ao passo que a SEMIA-587 e SEMIA-5019 inoculadas no genótipo Bossier expressaram alta frequência de nódulos com baixa atividade, mas de grande tamanho, no período de florescimento. Isto indicou que o grupo de maturação a que pertence o genótipo poderia estar influindo no ciclo de nodulação da estirpe. A última etapa, também conduzida em casa de vegetação, constou do estudo da interação entre dois isolados da estirpe SEMIA-587 e três genótipos selecionados em campo (Bossier, Planalto e SOC 81-76, do grupo precoce e IAC-9, Tropical e GO 79-1084, do grupo tardio). Foi avaliada a nodulação (número, peso e tamanho de nódulos e ARA) e rendimento da planta (peso da parte aérea e N-total). A ausência de interação significativa entre os tratamentos permitiu o estudo da ação independente da bactéria e da planta na simbiose, para todos os caracteres avaliados. Os resultados indicaram que somente a seleção praticada dentro da estirpe produziu acréscimos significativos, indicando a necessidade de pesquisas dirigidas à manipulação da planta hospedeira para aumento da eficiência simbiótica na fixação do N

Evaluation of soybean (Glycine max(L.) Merrill) genotypes and Bradyhizobium japanicum strains for their potential for symbiotic efficiency for N2 fixation

The present research was undertaken in three experimental phases. The first one, consisted on a field evaluation of 90 soybean genotypes, 68 of them from the early maturity group and 22 from the late maturity group, to measure the range of variability for nodulation ability in the host. The inoculation was performed with a mixture of three strains of B. japonicum: SEMIA-587, SEMIA-5019 and CB-1809. The parameters studied were: nodule number, weight, nitrogenase activity (ARA) and seed yield. The statistical anaIysis of data include da multidimensional analysis. The results revealed differences among genotypes, indicating their genetical potential for biological nitrogen fixation. It was not detected correlation between nodulation and seed yield. The second experimental phase was conducted under greenhouse conditions with the aim of evaluating the bacterial response within and among the three strains of B. japonicum when applied alone to Bossier (early maturity) and IAC-9 (late maturity) genotypes. Data were grouped in classes of frequencies of ARA and size of 60 individual nodules. The results showed the differencial behaviour among strains for bot parameters, and was atributed to the intrinsec genetical variation of the strain. Strain CB-1809 presented a normal distribution of data, whereas S EMIA-587 and SEMIA-5019 when inoculated in the Bossier, expressed a high frequency of nodules with low activity but with 1 arge size at the mid-flowering period. This indicated the nodulation traits of the strain probably was influenced by the lenght period for the genotype to reach the maturity. The last phase was also undertaken at the greenhouse and consisted in the study of the interaction among isolates from strain SEMIA-587 nodules picked up from one plant at the second phase and introduced into genotypes selected from the field trial - Bossier, Planalto and SOC 81-76 (early maturity group) and IAC-9, Tropical and GO 79-1084 (late maturity group). Nodulation (number, weight and size of nodules and NA) and weight and N-accumulation in the plant were also evaluated. The absence of a significant interaction among treatments showed that both plant and bacteria acted independently. The results indicated an overall superioty of the strain to enhance the N and weight of the plant. This suggested the need of research toward the manipulation of the host plant for enhancing the symbiotic efficiency of biological N2 fixation
