O objetivo do presente estudo foi caracterizar implantes de Progesterona e suas reutilizações e avaliar sua eficiência. Três experimentos foram conduzidos: para avaliação das quantidades de P4 liberadas (Experimento in vitro), teores plasmáticos proporcionados (Experimento in vivo - vacas ovariectomizadas) e taxas de concepção na IATF (Experimento a campo vacas paridas). No experimento in vitro, as três reutilizações do implante foram testadas em dissolutor de comprimidos com seis cubas, tendo uma solução álcool/água (60:40) como meio de difusão. Dois ensaios foram realizados, totalizando 12 implantes, sendo que no terceiro uso do dispositivo foi adicionado um anel de P4 (0,1g) e no quarto uso foram adicionados 2 anéis (0,1g cada). Os implantes novos foram testados em quadruplicata em um terceiro ensaio, com a mesma metodologia. Amostras foram coletadas nos tempos 2 min, 2, 4, 8, 12, 24, 48, 72 e 96h. As quantidades de P4 foram dosadas por Cromatografia Líquida de Alto Desempenho (HPLC). Para quantidade acumulada de P4, segundo dispositivo, em função do tempo, houve interação tempo*tratamento (P=0,0002). A quantidade de P4 diferiu (P=0,0152) conforme os subseqüentes usos dos implantes Cronipres® e também em relação ao tempo (P<0.0001). As equações que descrevem as quantidades liberadas são: dispositivos novos y = 240,69 + 84,127A x (R2=0,9721); 2°uso y = 75,918 + 42,595B x (R2=0,8858); 3°uso - y=62,94 + 31,29C x (R2=0,8629) e 4° uso y = 40,595 + 32,148C x (R2=0,7205). No experimento in vivo, cada uso foi testado em triplicata, totalizando 12 animais. Os implantes foram inseridos e amostras de sangue foram colhidas nos tempos 0 (momento da colocação do implante), 2, 4, 6, 8, 10, 12, 24, 48, 72, 96, 120, 144, 168 e 192h. As dosagens de P4 foram realizadas por Rádio-Imuno-Ensaio RIE, com kit comercial da DPC (Diagnostics Products Corporation, EUA). A AUC diferiu entre o primeiro e segundo (p<0,0001), segundo e terceiro (p=0,0010), mas não entre o terceiro e o quarto usos (p=0,8764). Para análise estatística destes experimentos, foram utilizados os PROC IML, GLM (comando REPEATED) e REG (teste in vitro, o qual gerou uma equação de regressão) ou avaliação da area under the curve AUC no teste in vivo, pelo teste t. No teste a campo, quatro lotes de vacas de corte paridas (n=763) Nelore e Simental x Nelore, com idade variando entre 3 e 10 anos e condição corporal entre 4 e 7 (escala de 1-9) foram utilizados. No D0 do tratamento todos os animais receberam 2 mg i.m. de Benzoato de Estradiol (Cronibest® - Biogénesis-Bagó, Argentina) e um implante Cronipres® (implante novo, contendo 1 g de P4 n=194; implante de segundo uso, n=190; implante de terceiro uso acrescido de um anel complementar de P4 (0,1g de P4), n=191; implante de quarto uso com um anel de P4 usado e dois anéis de P4 novos, n=188). No D8 procedeu-se a retirada do implante e aplicação i.m. de 300 UI de eCG (Novormon®, ScheringPlough, EUA) e 150 µg i.m. de D-cloprostenol (Croniben®, Biogénesis-Bagó, Argentina). No D9, todos os animais receberam 1mg i.m. de Benzoato de estradiol (Cronibest®) e, após 30h, foi realizada a IATF. Em todos os lotes, os tratamentos foram igualmente distribuídos. Os dados foram submetidos à regressão com análise de variância (PROC GLIMMIX do SAS Statistical Analysis Sistem®), separando-se como causas de variação: tratamento (implante novo, 2° uso, 3° uso e 4° uso), escore de condição corporal (ECC<5, ECC5 e ECC>5), ciclicidade (ciclando e nãociclando), lote (1 a 4), período pós parto (<3 meses, 3 a 5 meses e >5meses) e grau de sangue (Nelore e Simental X Nelore) e suas interações, eliminando-se as variáveis do modelo quando estas apresentavam p >0.2. No modelo final permaneceram as variáveis: tratamento (p=0,18), grau de sangue (p=0,11) e lote (p=0,14). Não houve diferença estatística nas TC segundo tratamento (implante novo 55,1%, 2° uso 47,9%, 3° uso 48,7%, 4° uso 44,15%) (p=0.18). A taxa de concepção geral foi de 49,1%.

The aim of this study was to characterize progesterone devices and their reuse and to evaluate its efficiency. Three experiments were conducted: to assess the quantities of released P4 (in vitro trial), plasma levels provided (in vivo trial -ovariectomized cows) and conception rates at FTAI (field trial suckled beef cows). In vitro trial, the three reuses of devices were tested in a tablet dissolutor with six tanks, with a solution of alcohol / water (60:40) as way of dissemination. Two trials were conducted, totaling 12 implants. The third use of the device was added with a P4 ring (0.1 g) and in the fourth use two rings were added (0.1 g each). The new implants were tested in quadruplicate in a third trial, using the same methodology. Samples were collected at 2 min, 2, 4, 8, 12, 24, 48, 72 and 96h. The amounts of P4 were measured by High Performance Liquid Chromatography (HPLC). For the cumulative amount of P4, there was a time * treatment interaction (P = 0.0002). The amount of P4 differed (P = 0.0152) as the subsequent use of implants Cronipres ® and also over time (P <0.0001). The equations that describe the amounts released are: new devices - y = 240.69 +84.127A x (R2 = 0.9721), second use - y = 75.918 +42.595B x (R2 = 0.8858), third use - y = 62.94 + 31.29C x (R2 = 0.8629) and fourth use - y = 40.595 + 32.148C x (R2 = 0.7205). In the in vivo trial, each use was tested in triplicate, totaling 12 animals. The implants were inserted and blood samples were collected at time 0 (time of implant insertion), 2, 4, 6, 8, 10, 12, 24, 48, 72, 96, 120, 144, 168 and 192h. The dosages of P4 were performed by Radio-Immuno-Assay RIA, using a commercial kit from DPC (Diagnostics Products Corporation, USA). The area under the curve differed between the first and second (p <0.0001), second and third (p = 0.0010) but not between the third and fourth use (p = 0.8764). For the statistical analysis of these experiments, the PROC IML, GLM (command REPEATED) and REG (for in vitro test, which generated a regression equation) or the t test (for the area under the curve assessment in vivo test), were used (Statistical Analysis System - SAS). In the field test, four groups of suckled Nellore and Simmental x Nellore beef cows (n = 763), aged between 3 and 10 years and body condition score between 4 and 7 (range 1-9) were used. On D0 of treatment all animals received 2 mg im of estradiol benzoate (Cronibest ® - Biogénesis-Bagó, Argentina) and an implant Cronipres ® (new implant containing 1 g of P4, n = 194; second use implant, n = 190; third use implant added with of an additional ring of P4 (0.1 g of P4), n = 191; fourth use implant added with a used ring of P4 and two new rings of P4, n = 188). On D8 the implant was removed and an i.m. application of 300 IU of eCG (Novormon ®, Schering-Plough, USA) and 150 mg i.m D-cloprostenol (Croniben ®, Biogénesis-Bagó, Argentina) was performed. On D9, all animals received 1 mg i.m. of estradiol benzoate (Cronibest ®) and after 30h, FTAI was performed. In all groups, the treatments were equally distributed. Data were subjected to regression with analysis of variance (PROC GLIMMIX in SAS ® Statistical Analysis System), separating as causes of variation: treatment (new use, second, third and fourth use), body condition score (BCS <5, ECC5 and BCS> 5), cyclicity (non-cycling and cycling), groups (1-4), postpartum period (<3 months, 3-5 months and> 5mounths) and breed (Nellore and Simmental x Nellore) and their interactions, eliminating the variables of the model when they showed p> 0.2. In the final model was consisted of treatment (p = 0.18), breed (p = 0.11) and group (p = 0.14). There was no statistical difference in conception rates between treatments (new device - 55.1%, second use - 47.9%, third use - 48.7% and fourth use - 44.15%) (p = 0.18). The overall conception rate was 49.1%.
