Utilizando--se a estirpe CB - 1809 de Rhizobium japonicum foram isolados mutantes resistentes a estreptomicina com o propósito de estudar o comportamento dos mesmos comparativamente a estirpe sensível original. Determinou-se a curva de sobrevivência da estirpe original frente a estreptomicina resultando que o modelo de resistência seguido foi o de “um só passo”, encontrando-se a CMI em torno de 10 - 16 mcg/ml de estreptomicina. Foram obtidas curvas de crescimento das linhagens sensível e resistentes a estreptomicina e através delas calculadas as taxas de crescimento das mesmas. O tempo de geração variou de 5 horas a 12 horas, havendo diminuição na taxa de crescimento dos mutantes resistentes. Crescimento em mistura de linhagens original e mutante resistente a estreptomicina mostrou que a porcentagem de mutantes caiu de 20.26% no início do cultivo para 7,47% ao final de 120 horas de cultivo. O valor da Meia Vida relativa (MVr) no caso foi de 77 horas. Inoculação de soja. em experimento de casa de vegetação, revelou que os mutantes perderam a capacidade de nodular, enquanto a estirpe original apresentou eficiência em fixar nitrogênio de ordem de 70% em relação as plantas adubadas com nitrogênio. A atividade da nitrogenase, medida em cromatógrafo de gás, foi em t orno de 35 mcMoles de N2 fixado/hora/vaso. Hipóteses para explicar a perda da capacidade nodulatória pelos mutantes resistentes, são discutidas.
Abstract in English
Streptomycin resistant mutants were obtained from the CB 1809 strain of Rhizobium japonicumwith the aim to study the behaviour of such mutants in comparison with the original strain. The survival curve of the original strain in the presence of streptomycin did show that the followed model was the “one step” resistant model. The MIC was around 10- 16 mcg/ml of streptomycin. The growth curves of original and streptomycin resistant strains were obtained and from them were determined the generation times for each studied strain. Generation times varied from 5 to 12 hours and the growth rates of resistant mutants were allways bellow of that of the original sensitiva strain. A mixed growth of a mutant strain and the original strain did show that the percentage of mutants decreased from 20,25% to 7,47% after 120 hours of mixed cultura. The value of the relative half life was 77 hours. Resistant mutant strains were not able to nodulate Gliune max. The original strain, on the other hand, did show efficiency in nitrogen fixation of about 70% in relation to plants which received nitrogen. The nitrogenase activity, measured by gas chromatrography was around 35 mcMoles of fixed N2 hour/vase. The possible causes of the nodulation inhability of the resistant mutants are discussed.
