A presente Dissertação trata de alguns aspectos do processo patológico causado pelo vírus da poliedrose nuclear (NPV) afetando duas espécies de Lepidoptera: Bombyx mori L. 1758 (Lep., Bombycidae) e Anticarsia gemmatalis Hübner, 1818 (Lep., Noctuidae), do dimensionamento das partículas infecciosas e incorporação de timidina tritiada no DNA viral. Através de fotomicrografias de cortes ultra e semifinos de células de corpo gorduroso células colunares do intestino médio e hemócitos de lagartas dos insetos doentes, constatou-se que o NPV estudado pode se desenvolver em quaisquer dessas células. Observações das fotomicrografias dos hemócitos em especial, apresentaram evidências sobre as vias pelas quais o NPV se sistematiza na lagarta. O aumento de volume dos núcleos de células do corpo gorduroso de lagartas de B. mori apresentando os primeiros sintomas da doença mostrou ser da ordem de 12,4 vezes maior que o de células sadias. A organização dos poliedros em hemócitos apresentou características com algumas diferenças em relação aos poliedros de outros tipos de células. Com respeito às dimensões das partículas de NPV de um mesmo inóculo afetando células colunares do intestino médio de lagartas de B. mori e A. gemmatalis assim como de partículas incluídas em poliedros e livres no nucleoplasma foram notadas diferenças estatisticamente significativas. As médias das larguras das partículas de NPV em B. mori (106,7 e 50,4 nm) foram maiores que as de A. gemmatalis (90,2 e 46,1 nm). As partículas de NPV no interior dos poliedros foram menores em comprimento e largura (270,9 e 43,3 nm) do que as partículas livres no nucleoplasma (300,2 e 47,3 nm) de células de B. mori. Os valores médios das partículas liberadas do poliedro em solução alcalina e observadas pelas técnicas de coloração negativa e metalização com platina foram 361 x 52 nm e 400 x 85 nm, respectivamente. O tratamento de poliedros em solução alcalina no período de 100 minutos provocou alterações das partículas que exibiram as membranas sem o nucleocapsídio interno. Partículas íntegras também foram observadas. A incorporação da timidina tritiada após 6 horas de tratamento medida através da densidade de grãos de prata/área revelou que o núcleo da célula afetada pelo NPV incorporou em média 1,51 gr/μm2 e o da célula sadia 0,30 gr/μm2 resultados estes diferindo significativamente. A incorporação da timidina tritiada no citoplasma de lagartas sadias e afetadas pelo NPV não apresentou diferenças significativas. Esses resultados indicam que a síntese do DNA viral se dá no núcleo da célula afetada pelo NPV.

This work takes in consideration the following: (i) some aspects of the pathologic process caused by nuclear polyhedrosisvirus(NPV) affecting two species of Lepidoptera: Bombyx mori L. 1758 (Lep., Bombycidae) and Anticarsia gemmatalis Hübner, 1818 (Lep., Noctuidae), (ii) size measurement of infectious particles; and (iii) incorporation of 3H-thymidine in the DNA of the virus. Ultrathin and thick photomicrographs sections of the fat body cells, the columnar cells of the midgut and the hemocytes of insects larvae infected with the virus showed that the studied NPV can develop in any of these cells. By use of the hemocyte photomicrographs the NPV development was observed in the larva. The volume increase of nuclei of fat body cells of B. mori larvae was about 12.4 times more in larvae showing initial disease symptoms than in the uninfected ones. The polyhedra arrangement in the hemocytes presented some characteristic differences in relation to the polyhedra of the other cell types. Regarding the dimensions of NPV particles of the same inoculum affecting the columnar cells of the midgut of B. mori and A. gemmatalis larvae, dimensions of particles enclosed within the polyhedra and free ones in the nucleoplasm, were found to be statistically different. Width and lenght means of NPV particles in B. mori (50,4 nm and 106.7 nm respectively were greater than those found in A. gemmatalis (46.1 nm and 90.2 nm). NPV particles of B. mori cells within the polyhedra with mean widths of 43.3 nm and lenght of 270.9 nm were smaller than those found in free state in the nucleoplasm whose mean widths and lenghts were 47.3 and 300.2 nm respectively. Mean dimension values of particles released from polyhedra by alkaline solution treatment and observed by uranyl acetate negative staining and platinum shadow casting techniques were 361 x 52 nm and 400 x 85 nm respectively. Polyhedra treatment in alkaline solution during 100 minutes, gave rise to changes in particles that exhibited membranes without their internal nucleocapsid. Whole particles were also observed. The take up of 3H-thymidine after treatment of 6 hours and measured by silver grain density (grains of silver/μm2 ) showed that the nucleus of NPV affected cells took up an average of 1.51 g/μm2 that those of uninfected cells only 0.30 g/μm2. The 3H-thymidine take up by the cytoplasms of healthy and NPV affected larvae did not present significant differences. These results show that the virus DNA synthesis takes place in the nucleus of NPV affected cells.
