Foram utilizadas atmosferas gasosas contendo 15N2 com baixo enriquecimento para testes de fixação simbiótica em feijão (Phaseolus vulgaris L.). Tanto nos testes de fixação em raízes noduladas como nos testes de fixação em plantas inteiras, nos quais as mesmas foram colocadas dentro de uma câmara de crescimento especialmente construída, os resultados obtidos foram positivos, e sugerem ser a metodologia usada bastante promissora para estudos mais detalhados sobre os vários aspectos da fixação simbiótica. Amostras de ar atmosférico foram purificadas absorvendo-se O2 e CO2 das mesmas por dois métodos, sendo o N2 assim obtido analisado e os resultados comparados. Foram analisados amostras de feijão coletadas em condições naturais com o intuito de se verificar a existência e a extensão da variação natural de 15N nas mesmas. Diversas amostras foram preparadas para análise por meio de 2 métodos, sendo feita a comparação entre os resultados obtidos. As amostras foram analisadas em um espectrômetro de massa Atlas-Varian modêlo CH-4. Os resultados são dados em δ0/00 de variação em relação a um gás de referência.
Abstract in English
Gaseous atmospheres containing 15N2 with low enrichment were used to test symbiotic nitrogen fixation in beans (Phaseolus vulgaris L.). The tests of fixation in nodulated roots and the tests of fixation in the whole plant, in which the plants were placed inside a especially constructed growth chamber, gave positive results and suggest that the methodology used can be very helpfull in more detailed studies on symbiotic fixation. Samples of atmospheric air were purified by absorption of O2 and CO2 by two methods. The purified N2 obtained was analysed and the results were compared. Samples of bean plant material were collected in natural conditions and analysed for 15N natural variation. Several samples were prepared for 15N isotopic analysis by two methods. The results obtained were compared. All samples were analysed in an Atlas-Varian Ch-4 model mass spectrometer, and the results were given in δ15N0/00 variation in relation to a standard gas.
