O presente trabalho teve como objetivo a determinação de parâmetros físicos e químicos que influenciam na atividade da enzima Redutase de Nitrato (NR) em plântulas de feijão. Esta enzima é uma das mais importantes no metabolismo nitrogenado da planta, pois ela representa o ponto inicial da assimilação do nitrogênio nítrico. Para os ensaios realizados, utilizamos sementes de Phaseolus vulgaris L., cvs. Goiano precoce e Carioca precoce, cedidas pela Seção de Radiogénetica do C.E.N.A. - Piracicaba-SP. Os experimentos foram conduzidos na casa de vegetação e nas câmaras de crescimento do Setor de Bioquímica de Plantas do C.E.N.A. Os resultados obtidos nos diferentes ensaios, demonstraram que a enzima é reduzida pelo substrato e possui um pH e temperatura ótimos para a incubação de 7,3 e 33°C respectivamente. As constantes de Michaelis-Menten também foram determinadas, sendo que para as folhas é de 6,8 x 10-3M e para as raízes de 9,7 x 10-4M . Verificou-se também que a atividade da NR varia com a idade da planta, tendo um máximo de atividade antes do florescimento dos cultivares utilizados. Também a atividade da NR varia de acordo com a posição da folha na planta, sendo que folhas sombreadas apresentaram menor atividade da enzima. A atividade da NR varia também com relação a idade da folha, sendo que as folhas mais novas possuem maior atividade. A luz também é um fator que afeta a atividade da NR. Plantas crescendo à luz quando foram colocadas em condições de escuro, tiveram sua atividade diminuída ao mínimo. Entretanto quando as plantas retornaram a luz, após 48 horas no escuro, tiveram a atividade da enzima restabelecida. Também o efeito de indutores e inibidores da NR foi estudado. A adição de intermediários da glicólise promoveu um aumento na atividade da enzima. Entretanto a adição de ácido isocítrico não teve efeito na indução ou inibição da enzima. Os reguladores de crescimento, ácido giberélico e 2-4-diclorofenoxiacético, promoveram um aumento da atividade da enzima. Também a auxina ácido indol acético, com o aumento da dose, promoveu um aumento da atividade enzimática. Com relação à adição dos aminoácidos ao meio indutor, foi verificado que os aminoácidos glicina e ácido glutâmico promoveram um acréscimo na atividade enzimática, enquanto que cisteína diminuiu sua atividade. Também a adição ao meio indutor de citrulina agmatina e putrescina, aumentou a atividade da enzima. Porém espermidina e espermina promoveram um decréscimo não significativo na atividade da NR. A irradiação com diferentes intensidades de radiação gama não teve efeito sobre a atividade da enzima, entretanto em relação a dose de radiação, houve uma inativação proporcional ao aumento da mesma. Estes resultados demonstram que a atividade da NR varia de acordo com as condições fisiológicas e ambientais em que a planta se encontra, tornando-se necessário para sua análise, estabelecer as condições ideais do ensaio.

The objective of this work is a determination of physic and chemical parameters that influence Nitrate Reductase activity in bean seedlings. This enzyme is perhaps more important than enzymes of the nitrogen metabolism, because it represents the initial point of the assimilation of Nitrate-N. Seeds of Phaseolus vulgaris L. cv. Goiano Precoce e Carioca Precoce, were supplied by the Secção de Radiogenêtica of CENA-Piracicaba-SP. The experiments were conducted in the greenhouse of the Setor de Bioquímica de Plantas do CENA. The data in the different assays demonstrate that the enzyme is induced by substrate and has single optimal pH and temperature for incubation of 7,3 and 33°C respectively. The MICHAELIS MENTEN constant for the nitrate was determined in the leaves (6,8 x 10-3M) and in the roots (9,7 x 10-4M). The activity of NR changes with the age of the plant, having maximal activity before flowering of the cultivars. The activity of NR also changes according to the position of the leaves in the plant, showing that the shaded leaves present low enzyme activity. The activity of NR varies with the age of the leaves and the young leaves have more activity than the others. Another parameter that regulated the enzyme activity is the light. Plants growing in the light, and then transferred to the dark, had the NR decreased. Furthermore, when the plants were returned to light, after 48 hours in the dark, the NR activity recovered. The effect of inducers and inhibitors of NR was also investigated. The addition of intermediates of glycolysis promoted and increase of NR activity. The addition of Isocitric Acid did not have any effect in the induction or inhibition of the enzyme. The growth regulators, Gibberellic Acid, and 2,4,D also promoted and increase of enzyme activity. The auxin Indol Acetic Acid, with increase of concentration, promoted an increase of enzyme activity. The addition of the aminoacids, glycine and Glutamic Acid, induced an increase of NR activity, and Cysteine decreased it. Also the addition of citrulin, agmatin and putrecin, to the induction medium, increased enzyme activity, Spermine and Spermidine did not increase the activity. However, irradiation with different intensities of Gamma irradiation, did not have any effect on enzyme activity. However, in relation to dose radiation with increase dosage there was a proportional inactivation of the enzyme. These results confirm that the NR activity varies according to the physiological and environmental conditions, that the plant and it was necessary to stablish the best conditions of assay the enzyme.
