O raquitismo da soqueira da cana-de-açúcar (RS) é uma das doenças mais importantes desta cultura, ocorrendo, se não em todas, na maioria das áreas de produção comercial do mundo. A falta de sintomas característicos tem dificultado uma avaliação rápida da intensidade do RS em campos comerciais e durante o melhoramento genético das variedades. Muitos métodos de diagnóstico são citados na literatura, porém não foram devidamente estudados quanto a sua eficiência e praticidade. Sendo assim, este trabalho se propôs a elucidar alguns pontos a fim de determinar um método que possua características desejáveis como praticidade na execução, economia de tempo, baixo custo e precisão. Concomitante a esta comparação de métodos, avaliou-se a resistência ao RS de algumas variedades comerciais de cana-de-açúcar para verificar a validade dos métodos na seleção de variedades resistentes. Três métodos foram avaliados: coloração do xilema através do fluxo transpiratório (CL) , observação da bactéria do RS em suco extraído do xilema (CT) e autofluorescência do metaxilema (FL). Verificou-se que os métodos (CL e CT) são equivalentes no diagnóstico do RS quando aplicados em plantas cujo processo de transporte de seiva bruta já sofreu interferência da bactéria. Nas condições testadas esta interferência ocorreu em torno de dez meses após a brotação. O método CT mostrou maior sensibilidade por detectar o patógeno em colmos na fase inicial do processo patológico. A simplicidade de execução e o baixo custo do método CL permitem a sua utilização para avaliação de campos comerciais, viveiros de mudas e seleção de variedades resistentes nos programas de melhoramento genético.

Comparison of methods for diagnosis of ratoon stunting disease of sugarcane (Clavibacter xyli subsp. xyli) and application on screening of resistant varieties

The ratoon stunting disease of sugarcane (RSD ) is one of the most important diseases of this crop, occurring, if not in alI, in most of the commercial production areas of the world. The lack of characteristic symptoms has made more difficult a quick evaluation of the intensity of RSD in commercial fields and during the breeding of the varieties. Many diagnostic methods are mentioned in the literature. However, they were not properly studied regarding their efficiency and practicability. Therefore, this work was proposed to illustrate some points, in order to determine a method that has desirable characteristics such as, practicability in execution, economy of time, low cost and accuracy. Simultaneously with this comparison of methods, the resistance to RSD was evaluated in some commercial varieties of sugarcane in order to verify the validity of the screening method of resistant varieties. Three methods were evaluated: staining of the xylem by means of the transpiratory flow (CL), observation of the RSD bacterium in juice extracted from the xylem (CT) and alkaline-induced metaxylem autofluorescence (FL). It was verified that the methods (CL and CT) are equivalent in diagnosis of RSD when applied in plants where the bacterium had already interferred in the transport of xylem sap. Under the conditions tested, this interference occurred in approximately ten months after sprouting. The CT method showed greater sensivity for detecting the disease in stalks in the initial phase of the pathological process. The simplicity of execution and the low cost of the CL method permit its utilization for evaluation of commercial fields and nurseries and screening of resistant varieties in the breeding programs
