Este estudo objetivou caracterizar e identificar uma bactéria que vem ocasionando lesões foliares em cana-de-açúcar. Os sintomas observados, em campo, são semelhantes àqueles descritos para a doença Estria Vermelha, causada por Pseudomonas rubrilineans. Todavia, as colônias bacterianas isoladas a partir de materiais com estes sintomas não corresponderam às características daquele patógeno. Foram, então, realizadas observações das características morfológicas, culturais, bioquímicas e fisiológicas das bactérias obtidas. A caracterização serológica foi feita através da técnica de dupla-difusão em ágar. Em condições de casa de vegetação, utilizando-se quatro isolados da bactéria em estudo e plantas de cana-de-açúcar, avaliou-se três técnicas de inoculação: deposição do inóculo no cartucho. infiltração foliar e infiltração no palmito. Os resultados mostraram que todos os métodos foram eficientes na reprodução de sintomas. As reações de resistência caracterizam-se por ausência de sintomas ou presença de pontos cloróticos isolados ou coalescentes, pequenas lesões necróticas avermelhadas circulares, circundadas por halos cloróticos podendo evoluir para lesões necróticas avermelhadas estreitas circundadas por regiões cloróticas; isoladas ou parcialmente coalescentes. Como reações de resistência moderada, foram consideradas a presença de pontos e lesões cloróticas estreitas e de lesões necróticas avermelhadas, estreitas e alongadas geralmente coalescentes mais longas e largas que as reações de resistência, podendo apresentar pontos ou pequenas áreas cloróticas associadas. As reações de suscetibilidade manifestaram-se pela coalescência de lesões necróticas avermelhadas, podendo atingir toda a largura do limbo foIiar, geralmente com a presença de áreas cloróticas com pontos avermelhados associados. Foi avaliado, em condições de casa de vegetação, o comportamento de plantas de cana-de-açúcar, quando inoculadas com Pseudomonas rubriIineans, Xanthomonas campestres pv. vasculorum e a bactéria em estudo. Os resultados mostraram que P. rubriIineans é mais exigente em umidade que as duas outras, necessitando, pelo menos, cinco dias de câmara úmida, após a inoculação p para o desenvolvimento das lesões. Não se observou diferenças significativas entre as reações das plantas às inoculações de X. c. pv. vasculorum e a bactéria em estudo. Foi determinada a patogenicidade de isolados destas duas bactérias em outras gramíneas de interesse econômico. Os dados obtidos mostraram que todos os isolados provocaram lesões foliares em plantas de milho, sorgo e aveia, mas não em plantas de trigo, em condições de casa de vegetação. A análise dos resultados dos experimentos realizados permitiram caracterizar e identificar esta bactéria como pertencente ao gênero Xanthomonas, espécie X. campestres, sugerindo tratar-se do patovar vasculorum.

Occurrence and characterization of Xanthomonas campestris in sugarcane (Saccharym), in the States of São Paulo and Paraná
Abstract in English
The characterization and identification of a bacterium which cause lesions in sugarcane leaves was the object of this study. Symptoms observed in the field were similar to those described as Red Stripe, disease caused by Pseudomonas rubrilineas. However, the bacterial colonies isolated from sugarcane tissues with those symptoms did note correspond to that pathogen. Observations about morphological, cultural, biochemical and serological characteristics were made. Sugarcane plants were evaluated by three inoculation methods: inoculation placed into the spindle, leaf infiltration with the bacterial suspension and injection of the suspension into the spindle. The results showed that all the methods were efficient in reproducing the symptoms. Differences in the types of resistance and susceptibility reactions were inoculated with bacterial isolates. The resistance reactions were characterized by the absence of visible symptoms or by presence of isolated or coalescent chlorotic dots, and small, reddish, circular necrotic lesions surrounded by chlorotic halos; these lesions may become narrow and elongated, surrounded by chlorotic areas usually isolated or partially coalescent. The moderate resistance reactions were characterized by presence of chlorotic areas containing reddish, circular necrotic lesions, or reddish, narrow and elongate necrotic lesions, usually coalescent, which are wide and longer than those observed on resistant plants and surrounded by chlorotic areas. The susceptibility reactions comprised the coalescence of reddish, elongated necrotic lesions that can reach all the width of the leaf, usually with red-flecked chlorotic stripes. The behavior of sugarcane plants inoculated with Pseudomonas rubrilineans, Xanthomonas campestris pv.. vasculorum and the bacterium studied was evaluated under greenhouse conditions. The results showed that Pseudomonas rubrilineans needs a period of high relative humidity longer than the two others, demanding at least 5 days in moist chamber, after inoculation in the reaction was observed among the plants inoculated with Xanthomonas campestris pv. vasculorum and with the bacterium in study. The pathogenicity of isolates of these two bacteria were established in other important crops of the “Gramineae” family. The data obtained under greenhouse conditions showed that all the isolates cause leaf lesions in corn, sorghum and oat plants but not in wheat plants. The analysis of the results allowed to characterize and identify this bacterium as belonging to genera Xanthomonas, species X. campestris. The possibility of belonging to the pathovar vasculorum could not be discarded.
