Doze isolados de Pythium foram obtidos de raízes de alface cultivada em sistemas hidropônicos comerciais, apresentando ou não sintomas de apodrecimento. Três desses isolados foram identificados como Pythium helicoides (H1, H2 e Hb), cinco como pertencentes ao grupo F (F1 a F5) e quatro ao grupo T (T1 a T4) de Pythium. A identificação das espécies foi realizada baseando-se na análise de características morfológicas. O efeito da temperatura (10, 15, 18, 21, 24, 27, 30, 37 e 40°C) sobre o crescimento micelial foi determinado para todos os isolados. As temperaturas mínima e máxima, estimadas pela função beta generalizada, variaram de 3,5 a 10°C e de 40 a 40,7°C, respectivamente. A temperatura ótima foi de 24 a 37°C para os isolados de P. helicoides, de 25 a 35°C para o isolado F4 e de 21 a 30°C para os demais isolados. O potencial patogênico dos isolados foi avaliado, inoculando-se sementes de alface semeadas em ágar-água, a 21 e 30°C. A 30°C, os isolados de P. helicoides foram notadamente os mais patogênicos, ocasionando 100% de mortalidade das sementes logo após sua germinação. A 21°C, todos os isolados induziram subdesenvolvimento de plântulas, acompanhado ou não de necrose dos tecidos radiculares. A sensibilidade dos isolados H1, F1 e T1 a nove fungicidas foi determinada "in vitro", nas concentrações de 0; 0,1; 1; 10 e 100 ppm de ingrediente ativo. De modo geral, mancozeb, metalaxyl + mancozeb, cymoxanil + maneb e cymoxanil + famoxadone apresentaram maior eficiência. O efeito de espalhantes adesivos e do fungicida fluazinam, nas concentrações de 25, 50 e 100 ppm do ingrediente ativo, sobre a integridade e motilidade dos zoósporos e sobre o crescimento micelial dos isolados H1, F1 e T1 foi também avaliado. Todos esses produtos ocasionaram a paralisação do movimento e, com exceção de Silwet L-77 AG® e Agrotensil® (a 25 ppm) e Tween 80® (25 a 100 ppm) provocaram lise dos zoósporos. O crescimento micelial foi pouco afetado pelos surfactantes testados. Porém, a 100 ppm, o desenvolvimento dos isolados F1e T1 foi completamente inibido pelo Espalhante adesivo Bayer® e o crescimento de F1, eliminado pelo Energic®. Hipoclorito de sódio, hipoclorito de cálcio e amônia quaternária foram testados na inibição dos isolados H1, F1 e T1 de Pythium, nas concentrações de 0; 0,1; 1; 10; 100; 1000 e 10.000. Esses desinfetantes mostraram-se ineficientes na erradicação de micélio e a completa inibição dos isolados apenas ocorreu com a utilização de altas dosagens, 1000 ppm para hipoclorito de sódio e de cálcio e 10.000 ppm para amônia quaternária.

Identification, pathogenicity and in vitro sensibility to chemical products of Pythium species from lettuce (Lactuca sativa L.) grown bydroponically

Twelve Pythium isolates were obtained from roots of lettuce grown hydroponically in commercial systems, showing or not symptoms of rotting. Three of them were identified as Pythium helicoides (H1 H2 and H3), whereas five were shown to belong to group F (F1-F5) and four to group T (T1-T4) of Pythium. The identification of the species was based on morphological characteristics. The effect of temperature (l0, 15, 18, 21, 24, 27, 30, 37 e 40°C) on the mycelial growth was determined for a1l isolates. Minimum and maximum temperatures, estimated by generalized beta function, varied from 3,5 to 10°C and 40 to 40,7°C, respectively. The optimum temperature ranged from 24 to 37°C for Pythium helicoides isolates, from 25 to 35°C for isolate F4 and 21 to 30°C for the remaining isolates. The pathogenic potential of the isolates was evaluated by the inoculation of lettuce seeds plated in water-agar, at 21 and 30°C. At 30°C, Pythium helicoides isolates were clearly the most pathogenic, determining 100 % seed mortality soon after germination. At 21°C, a1l isolates induced reduction of seedling growth, associated or not with root tissue necrosis. Sensibility of isolates H1 F1 and T1 to nine fungicides was determined "in vitro", at concentrations of 0; 0,1; 1; 10 and 100 ppm of active ingredient. Mancozeb, meta1axyl + mancozeb, cymoxanil + maneb and cymoxanil + famoxadone showed, in general, the highest efficiency. The effect of surfactants and fluazinam, at the concentrations of 25, 50 and 100 ppm of active ingredient, on the integrity and motility of zoospores and mycelial growth of isolates H1, F1 and T1 was also evaluated. All these products determined movement paralyzation and with the exception of Silwet L-77 AG®; and Agrotensil® (at 25 ppm) and Tween 80® (at 25 to 100 ppm), they induced zoospore lysis. Mycelial growth was little affected by the surfactants tested. At 100 ppm, however, growth of isolates F1 and T1 was completely inhibited by Espalhante adesivo Bayer® and the growth of F1, prevented by Energic®. Sodium hypochlorite, calcium hypochlorite and quaternary ammonium were tested against isolates H1, F1 and T1 of Pythium, at the concentrations of 0; 0,1; 1; 10; 100; 1000 and 10.000 ppm. These disinfectants revealed no efficiency in eradicating the mycelium and the complete inhibition of the isolates occurred only at high doses, 1000 ppm for sodium and calcium hypochlorites and 10.000 ppm for quaternary ammonium.
