Visando avaliar o potencial do microorganismo Saccharomyces cerevisiae em proteger plantas de café (Coffea arabica L. cv. Mundo Novo) contra o patógeno Hemileia vastatrix, plantas de um ano de idade foram aspergidas com uma suspensão de células vivas ou autoclavadas da levedura, antes, simultaneamente ou depois da inoculação com o patógeno. O efeito protetor de diferentes concentrações de S. cerevisiae, bem como a duração de tal efeito, também foram estudados. Os efeitos do tratamento com suspensão de células vivas ou autoclavadas da levedura e seus respectivos filtrados sobre a germinação, formação de apressórios e persistência dos urediniosporos do patógeno na superfície foliar foram observados por microscopia óptica ou de fluorescência. Os resultados mostraram que a proteção de plantas de café por S. cerevisiae só se manifesta nos tratamentos realizados antes ou simultaneamente à inoculação com H. vastatrix. Nenhuma proteção foi observada nos tratamentos realizados depois da inoculação com o patógeno. A proteção observada é local, não existindo uma resposta de proteção sistêmica. O maior efeito de proteção da levedura foi observado para concentrações de 20 mg/ml ou maiores, ficando ao redor de 70% de proteção em relação ao controle. A proteção se mantém nestes níveis no período que vai dos 4 dias a 2 semanas após o tratamento com a levedura, decrescendo gradativamente depois disto. Nos tratamentos onde foram utilizadas células vivas ou autoclavadas da levedura, verificou-se uma inibição significativa da germinação "in vi tro" e "in vivo", bem como um decréscimo acentuado do número de urediniosporos do patógeno presentes na superfície das folhas de café. Paralelamente estes são os tratamentos que mostraram uma efetiva proteção das plantas à ferrugem. Filtrados de células vivas ou autoclavadas da levedura mostraram um efeito pouco significativo sobre a germinação dos urediniosporos, assim como sobre a permanência destes nas folhas, sendo que tais filtrados não foram eficientes em proteger as folhas de café contra ferrugem. Suspensão de células ou filtrados da levedura não afetaram a formação de apressórios do patógeno. Os resultados sugerem que a proteção à ferrugem observada quando plantas de café são tratadas com S. cerevisiae, é devida basicamente a um efeito de controle biológico do patógeno pela levedura, podendo este efeito ser dividido em dois mecanismos distintos: o primeiro seria responsável por uma diminuição do número de urediniosporos do patógeno que permanecem na superfície das folhas e o outro por uma inibição da germinação dos urediniosporos restantes, provavelmente por antibiose. Nenhuma evidência foi verificada para sugerir a hipótese do envolvimento de um mecanismo de indução de resistência na proteção observada.

Protection of coffee plants (Coffea arabica L.) against Hemileia vastatrix Berk. et Br. by Saccharomyces cerevisiae Meyen

In order to evaluate the potential capacity of Saccharomyces cerevisiae to protect coffee plants (Coffea arabica cv. "Mundo Novo") against the pathogen Hemileia vastatrix, one year-old plants were sprayed with a suspension of either living or autoclaved cells of S. cerevisiae before, simultaneously or after the challenger inoculation with the pathogen. The protective effect of increasing concentrations of S. cerevisiae as well as the duration of such effect were also studied. The effects of the treatment with living or autoclaved yeast cells and theirs respective filtrates in the urediniospores, appressorium formation and persistence of the urediniospores of the pathogen on the leaf surface were observed under optical or fluorescence microscopy. The results showed that protection of coffee plants by S. cerevisiae is only expressed in treatments where protectant was applied before or simultaneously with the pathogen inoculation. No protection was observed in the treatments performed after the pathogen inoculation. Protection is locally evidenced and no systemic response was detected. The highest effectiveness of the protectant was observed for concentrations of 20 mg/ml or highers, with a protection of at about 70% of the control. The protection persisted in this highest level during a period of time between 4 days and 2 weeks after the yeast treatment, decreasing gradually after that. In treatments where living or autoclaved yeast cells were used, it was observed a significative "in vitro" and "in vivo" inhibition of germination of the uredeniospores of the pathogen as well as a marked decrease in the number of spores present on the coffee leaves surface. These treatments also corresponded to the highest effectiveness in protecting plants against coffee leaf rust. Living and autoclaved yeast cells filtrates showed only a slight effect on the urediniospores germination and in their persistence on the leaf surface. Yet, the mentioned filtrates were unable to protect coffee leaves against the pathogen. The appressoria formation was not affected neither by yeast cell suspensions nor by their filtrates. The results strongly suggest that the protection against coffee leaf rust observed when plants were treated with S. cerevisiae is essentially due to a biological control effect by the protectant and that there are two distincts mechanisms involved; one that leads to a decrease in the number of urediniospores present on the leaf surface and the other, that inhibits the germination of the remainder urediniospores probably by antibiosis. No evidences were observed to support the hypothesis of an induced protection mechanism involved in the protective effect.
