O desenvolvimento do fungo Hemileia vastatrix Berk et Br. raça II predominantemente, foi observado ao microscópio de epifluorescência, até 288hs após a inoculação em Coffea arábica c.v. "Mundo Novo" com ou sem resistência induzida pela aplicação prévia de extrato de fermento (MN-R e MN, respectivamente), e no híbrido "Sarchimor" (Sch). O fungo se desenvolveu de forma semelhante nas três interações estudadas, até a diferenciação de vesícula sub-estomática que ocorreu entre 24 e 36hs após a inoculação. A expressão da incompatibilidade entre cafeeiro e ferrugem nas interações Sch X raça II e MN-R X raça II foi detectada ao microscópio 48-72hs após a inoculação do fungo, quando a inibição do desenvolvimento do fungo pôde ser notada. A grande maioria das colônias foi abortada após a vesícula sub-estomática, no estágio de 1 célula-mãe de haustório (1 CMH) em Sch e 2 CMH em MN-R. Na interação Sch X raça II, 192hs após a inoculação do fungo, ca. de 50% das colônias apresentavam-se associadas a células epidérmicas ou mesofílicas com sinais de necrosamento. As estruturas do fungo nas colônias abortadas exibiam uma auto-fluorescência amarela semelhante a das células necrosadas do hospedeiro que as circundavam. Nessa interação, aparentemente a morte do fungo precedeu á reação de necrosamento das células hospedeiras. Na interação MN-R X raça II, 80% das colônias não puderam mais ser visualizadas pelas técnicas utilizadas, 72hs após a inoculação do fungo, sugerindo ter ocorrido profundas alterações nas estruturas do fungo que não foram acompanhadas de reação de necrosamento das células hospedeiras. Em todas as três interações estudadas, a maioria das primeiras CMH diferenciou haustório em célula epidérmica entre 48 e 72hs após a inoculação. Na combinação incompatível Sch X raça II os primeiros haustórios mostraram um crescimento reduzido, e 96hs após a inoculação a grande maioria apresentou-se encapsulada em um espessamento, que aparentemente se desenvolve a partir da parede celular do hospedeiro. Os resultados obtidos sugerem que a expressão de resistência em Sch ocorre mais rapidamente do que em MN-R, e através de mecanismos distintos.

Sequence of primary events of Hemileia vastatrix development in coffee plants with genetic susceptibility, resistance or genetic resistance induced
Abstract in English
Development of Hemileia vastatrix Berk. et Br., predominantly race II, on coffee leaves was observed by epifluorescence microscopy for 288hs from the time of inoculation of plants. Three types of plants were used, Coffea Arabica cv. "Mundo" Novo" (MN), "Mundo Novo" plants that had previously been induced to express resistance by prior treatment with yeast fitrate (MN-R), and the coffee hybrid of C. Arabica X C. canephora, “Sarchimor” (Sch). Fungal growth was similar in the three interactions studied up to the time of substomatal vesicle differentiation, which occurred between 24 and 36hs after inoculation. The expression of incompatibility in Sch X race II and MN-R X race II interactions was detected 48-72hs after inoculation, when inhibition of fungus development was observed. The majority of colonies was aborted after substomatal vesicle differentiation. In Sch this occurred at 1 haustorial mother cell (HMC) stage whereas in MN-R colony abortion occurred at the 2HMC stage. In the Sch X race II interaction, 192hs after fungus inoculation, 50% of the colonies were associated with necrotic host cells of the epidermis or of the mesophyll. Fungus structures of the aborted colonies were collapsed, and exhibited a yellow auto-fluorescence like the surrounding necrotic host cells. In hybrid Sch, death of the fungus apparently occurred before the necrosis of the host cells. In the MN-R X race II interaction, 80% of the colonies could not be visualized by 72hs after inoculation. This suggested that profound alterations must have occurred in the fungus even though necrosis of host cells did not occur in these infection sites. In all three interactions studied, the majority of the first HMCs differentiated one haustorium in the epidermal cell. In the Sch X race II combination, the first haustorium had reduced growth and the majority was enveloped by a sheath which appeared to grow up from the host cell wall.
