Effects of Meloidogyne incognita race 3 and M. javanica on the absorption and translocation of nitrogen, phosphorus and calcium and on carbon partitioning in cultivars of soybean
Abstract in English
The effects of the nematodes Meloidogyne incognita race 3 and M. javanica on the absorption and translocation of N, P and Ca were studied in two soybean cultivars differing in their susceptibility to the pathogens. ln three independent experiments, after 40 days from inoculation, plants infected with four levels of inoculum (0, 3,000, 9,000 and 27,000 eggs) were supplied with 15N, 45Ca and 32P. The experiments were carried out in a glasshouse. The plants were grown in plastic pots with the bottom removed and replaced with a net such that the roots emerged below in a second pot maintained partially filled with water. The roots of the upper pot were inoculated with nematode eggs and labelled nutrients were supplied to the lower pot with a diluted nutrient solution. After a period for absorption, the roots protruding in the lower pot were discarded and the roots of the first pot and the shoot were collected for analysis. lnfected plants had higher root dry mass and lower shoot dry mass. Data for the whole plant showed that infected and non infected plants had similar amounts of endogenous N and Ca, but less P, indicating a lower absorption for the latter nutrient. On the other hand, the endogenous content in the shoot of infected plants showed that the three nutrients were poorly translocated compared with non infected ones. The analysis of the labelled elements supplied 40 days after inoculation showed that absorption and translocation of 15N was not affected but infected plants absorbed and translocated less 45Ca and 32P to the shoot. No significant difference was observed regarding nematode species or plant cultivars used. To study the effects of nematodes on carbon partitioning in soybean plants, the same cultivars, nematodes and infection levels were used. After 30 days from inoculation, the plants were exposed to 14CO2 for four hours and after 24 h the plants were harvested for analysis. ln the roots of both cultivars M. javanica produced more eggs than M. incognita race 3. Increase of root fresh weight and reduction of shoot dry weight was observed with increase of inoculum level. ln roots, there was an increase in the total radioactivity and specific radioactivity in sucrose and reducing sugars with the increase of the inoculum level for both nematodes. Highest specific radioactivity was observed in reducing sugars, despite the endogenous content of sucrose being higher than reducing sugars. The results show that nematodes are strong metabolic sinks, causing marked alterations in the pattern of carbon partitioning in infected soybean. It was also observed that this pattern can be different according to the nematode species and soybean cultivar combination.
