O nematóide de galhas da espécie Meloidogyne javanica é um patógeno de importância econômica na cultura da soja. O controle eficiente deste nematóide é na maioria das vezes, dependente da utilização de genótipos de soja resistente. A interação genética deste patossistema é complexa e a análise da expressão simultânea de genes envolvidos no processo infeccioso se faz necessária. O objetivo deste trabalho incluiu a identificação e caracterização de genes envolvidos na interação soja-nematóide por comparação das diferenças na expressão gênica ocorridas em genótipos de soja resistente (JF 7056) e suscetível (266-S) a M. javanica, por meio de microarranjos de DNA. Estes genótipos foram utilizados para a obtenção de quatro tratamentos: plantas resistentes inoculadas, não inoculadas, plantas suscetível inoculadas e não inoculadas com uma concentração de 6700 juvenis J2 de M. javanica. O RNA total foi extraído das raízes de cada tratamento e utilizado para construir bibliotecas de cDNA. Após o seqüenciamento, aproximadamente 1000 seqüências únicas foram amplificadas e fixadas em lâminas de vidro. As sondas de cDNA foram marcadas com Cy3 (plantas inoculada) e Cy5 (plantas não inoculadas) sendo estes fluoróforos incorporados diretamente por transcrição reversa. Os arranjos foram hibridizados e intensidade da imagem foi quantificada, normalizada e analisada através da ferramenta estatística SAM. A comparação dos padrões de expressão nas plantas resistentes e suscetíveis mostrou que no genótipo resistente JF 7056, 208 genes apresentam expressão diferencial. Entre eles, 158 genes foram induzidos e 50 genes foram reprimidos nas plantas resistentes a M. javanica. No genótipo suscetível 266-S, 103 genes apresentaram expressão diferencial, onde 76 genes foram induzidos e 27 foram reprimidos nas plantas suscetíveis a M. javanica. Estes genes foram distribuídos em diferentes categorias funcionais atribuídas por comparação de seqüências de outros organismos depositados em banco de dados. Entretanto, grande proporção dos genes expressos não apresentou homologia com outros genes conhecidos sendo provavelmente, específicos em soja. A expressão induzida nas plantas resistentes inoculadas está relacionada em sua maioria com genes que atuam na interação patógeno-hospedeiro. Entre os genes com expressão induzida no genótipo resistente está Hs1pro-1, (primeiro gene de Resistência a nematóides identificado em plantas) e PR proteína SH2 (relacionada a patogenicidade). A expressão induzida nas plantas suscetíveis inoculadas, no entanto, apresentou vários genes envolvidos com a produção de hormônios como etileno, auxina, giberelina e proteínas relacionadas à maturação e senescência em plantas

Genetic expression of soybean Meloidogyne javanica interaction by DNA microarrays

The root-knot nematode, Meloidogyne javanica is an economically important pathogen of soybeans. Effective control of the nematode is often dependent on the use of resistant genotypes. The genetic interactions are complex and the simultaneous analysis in expression genes involved in the infectious process is done necessary. The use of DNA Microarray technology promises to accelerate rapidly our understanding of infectious disease processes, by allowing the examination of thousands of genes and gene products. To explore the plant genes putatively in the events of nematode-plant parasitism, was used DNA Microarray to conduct expression profiling of the early stages of infection of soybean genotypes. The objective of this work included the identification and characterization of genes involved in the interaction soybean-nematodes for comparison of the differences in the genetic expression. The infection experiments were conducted with four treatments: genotype of resistant soybean (JF 7056) inoculated and non-inoculated, genotype of susceptible soybean (266-S) inoculated and non-inoculated with a concentration of 6700 juvenile J2 of M. javanica RNA was extracted from the roots of each treatment and used to build cDNA libraries. After the sequencing, approximately 1000 unique sequences of DNA were amplified and spotted in glass slides. The cDNA probe was labeled with Cy3 (inoculated plant) and Cy5 (non inoculated plant), both incorporate directly during reverse transcription. The arrays were hybridized and intensity of the image was quantified, normalized and analyzed through the statistical tool SAM. The comparison of the expression patterns in the resistant and susceptible plants showed that in the genotype JF7056, 208 genes present differential expression. Among them, 158 genes were induced and 50 genes were repressed in the resistant plants to M. javanica. In the genotype 266-S, 103 genes presented differential expression, where 76 genes were induced and 27 were repressed in the susceptible plants to M. javanica. These genes were distributed in different functional categories attributed by comparison of sequences of other organisms deposited in database. However, great proportion of the genes expressed didn't present homology being probably, specific to soybean. The expression induced in the inoculated resistant plants is related in majority with genes that act in the host-pathogen interaction. Among the genes with expression induced in the resistant genotype are Hs1pro-1, (first Resistance gene identified in plant nematode) and PR protein SH2 (Pathogenesis related). The expression induced in the inoculated susceptible plants, however, it presented several genes involved with the production of hormones as ethylene, auxin, gibberellin, senescence and maturation related protein
