“Plantlets” de Eucalyptus urophylla, de E. grandis e de um híbrido (E. grandis x E. urophylla) e plântulas de E. urophylla e de E. grandis foram inoculadas in vitro com micélio de seis isolados de Pisolithus tinctorius, em substrato de turfa-vermiculita umedecido com solução nutritiva, com e sem carbono orgânico disponível. Em "plantlets" do híbrido e de E. urophylla, a colonização ectomicorrízica foi no mínimo de 82,6% e 66,1% do sistema radicular, respectivamente, com pouca variação entre os isolados fúngicos, após 45 dias de incubação no substrato com carbono orgânico disponível. Em "plantlets" de E. grandis, a colonização foi no máximo de 56,2%, com grande variação entre os isolados fúngicos. Em plântulas, ela foi de pelo menos 91,1% com pequena variação entre os isolados fúngicos e entre espécies hospedeiras. Na ausência de carbono orgânico, observou-se colonização ectomicorrízica de pelo menos 92,3% e 60,0% para um isolado de P. tinctorius em plântulas de E. urophylla e E. urophylla, respectivamente e no máximo 15,0% e 8,0% em E. urophylla e E. grandis, respectivamente, para outros dois isolados fúngicos. Os demais isolados permaneceram num nível intermediário. Em "plantlets" com elevada colonização ectomicorrízica formada em meio orgânico e transferidas para solo previamente esterilizado, somente para três isolados fúngicos ocorreu perpetuação das ectomicorrizas, com no máximo 10% do sistema radicular colonizado, 90 dias após o transplante. Em plântulas com elevada colonização ectomicorrízica e transferidas para o solo previamente esterilizado, a perpetuação das ectomicorrízicas foi constatada em grande quantidade somente para aquelas formadas na ausência de nutrientes orgânicos, com grandes diferenças entre a aagressividade dos isolados fúngicos em cada espécie hospedeira. Eucalyptus urophylla apresentou colonização micorrízica bem maior, do que E. grandis, 90 dias após o transplante.

Plantlets of Eucalyptus urophylla, E. grandis and (E. grandis x E. urophylla) hybrid and seedlings of E. urophylla and E. grandis were inoculated in vitro with mycelium of six isolates of Pisolithus tinctorius in a peat-vermiculite substrate moistened with a nutrient solution, with and without available organic carbon. Ectomycorrhizal colonization was at least 82,6 and 66, 1% of the root system in the hybrid and E. urophylla plantlets, respectively, after an incubation period days in the presence of organic carbon. There was of 45 little variation among fungal isolates. For E. grandis, the maximum colonization rate was 56,2%, with great variation among isolates. Root colonization in seedlings was a least 91,1%, with little variation among host materials and isolates of Pisolithus tinctorius. In the absence of organic carbon in the substrate, the ectomycorrhizal colonization for one of the isolates was at least 92,3 and 60,0% in seedlings of E. urophylla and E. grandis, respectively. For two other isolates maximum colonizations were 15,0 and 80% in E. urophylla and E. grandis, respectively. The other three isolates gave colonization rates. Perpetuation of ectomycorrhizal development 90 days after transplant to sterilized soil of plantlets with high ectomycorrhizal colonization in the organic substrate was obtained only for three isolates of the fungus at a maximum rate of 10% of the root system. In seedlings, perpetuation of ectomycorrhizal development after transplant was successfully obtained only for those that were formed in the absence of organic carbon. In this case, there were great differences among isolates in each host material. E. urophylla showed greater ectomycorrhizal development than E. grandis.
