Procurou-se uma metodologia para testes de resistência varietal de cana-de-açúcar a Pseudomonas rubrilineans(Lee et al) Stapp em casa-de-vegetação. As plantas foram inoculadas com a idade de 30 dias e as avaliações foram feitas 21 dias depois da inoculação. Os testes foram realizados com plantas resistentes e suscetíveis crescendo vigorosamente e os resultados analisados estatisticamente. Em todos os experimentos, durante os dias de câmara úmida, a umidade relativa foi mantida no intervalo de 90 a 100%. Após este período, as plantas ficaram em condições normais de casa-de-vegetação. Em cada experimento foi mantido um controle parcial da temperatura. Foram comparados três métodos de inoculação: injeção, pulverização e gotas no cartucho foliar. Foi selecionado o método de injeção como o mais eficiente, capaz de reproduzir todos os sintomas observados no campo. Comparou-se três concentrações de inóculo: 3x104; 3x106 e 3x108 talos bacterianos/ml. A concentração de 3x106 apresentou os melhores resultados. Observou-se os efeitos de três períodos de câmara úmida três, cinco e sete dias. Constatou-se que o período de 5 dias foi suficiente para indução de todos os sintomas nas plantas inoculadas.

In this work, the establishment of a method for testing varietal resistence to Pseudomonas rubrilineans (Lee et al) Stapp in sugar cane growing in a green house was tried. Plants were inoculated when they were 30 days old and evaluations were realised 21 days later. Tests were done on resistant and susceptible plants which were in their vigorous growing phase and the results obtained were analysed statistically. In all the experiments, when the plants were growing in a humid chamber the relative humidity was maintained at 90-100%; afterwards the plants were left in the prevailing conditions of the greenhouse. In each experiment there was a control maintained under partially controlled temperatures. Three methods of inoculating were compared: injection, spraying and putting drops of the inoculum in the whorl. The injecting method was found to be the most efficient for the reproduction of all the symptoms commonly observed in the field. Three concentrations of the inoculum were compared: 3x104, 3x106 and 3x108 bacterial cells/ml. A concentration of 3x106 gave the best results of all the 3. As for the effects of keeping the plants in a humid chamber for 3, 5 and 7 days on the inducing symptoms in the host, a period of 5 days was found to be sufficient for the purpose.
