Determinou-se a curva de crescimento in vitro Xanthomonas albilineans em meio de Wilbrink modificado, a 28°C e no escuro; seu patamar de crescimento foi de 7,5 a 8,5 x 108 UFC/ml, o que corresponde a uma densidade ótica de 0,15 a 0,20 no comprimento de onda de 625 nm, e 132 a 144 horas de incubação; a adição de 20% de extrato de células mortas de X. albilineans ao meio de Wilbrink modificado, propicia diminuição do período de "lag-fase" porém o número de células viáveis passa a ser menor. Estudando-se o seu comportamento quando exposta a diferentes regimes de temperatura, verificou-se que o crescimento in vitro a 40°C é praticamente nulo, porém a patogenicidade é mantida por até 96 horas; a taxa de crescimento diminui mas a patogenicidade não é perdida quando se faz alternância de 28°C e 40°C e mantém-se por até 72 horas quando muda-se de 28°C para 40°C, apesar de, neste caso, o crescimento ser paralisado. Testando-se o milho doce Cuba como planta indicadora para a escaldadura das folhas da cana-de-açúcar, verificou-se que o mesmo acusa sintomas quando inoculado com caldo de colmos que pelo menos apresentem descoloração vascular, sendo mais fácil quando o inóculo é de variedades suscetíveis, que não apresentem infecção latente; em cultura pura, apenas algumas células bacterianas inoculadas por injeção no palmito foliar do milho doce Cuba já induzem à manifestação de escaldadura, cujos sintomas foliares são influenciados pelas condições de luminosidade e a descoloração vascular pela presença de outros microorganismos como Fusarium moniliforme e Erwinia herbicola. O milho doce Cuba, porém, pode ser usado como planta-teste em qualquer fase de seu desenvolvimento, sendo um método prático e relativamente seguro. O milho doce Cuba propiciou a descoberta de um possível modo natural de disseminação da bactéria, pelas gotículas de gutação, pois acusou sintomas quando inoculado com estas gotículas retiradas de folhas infectadas de cana-de-açúcar, sugerindo que a escaldadura pode se transmitir no campo sem a interferência do homem. Testando-se a possibilidade de cura desta doença, constatou-se que termoterapia in vivo por até 40 dias a 40°C não foi um método eficiente. A associação de termoterapia in vivo e a cultura de ápices meristemáticos, propiciou altas porcentagens de cura. Porém, mesmo com aplicação de giberelina para maior alongamento dos colmos, e exposição das plantas à luz contínua para aumentar a resistência, houve aparecimento de escaldadura em alguns "explants" obtidos. Já que para esta doença não existe cura natural em variedades suscetíveis e o tratamento térmico a 50,5°C/2 horas também não é eficaz, sugere-se que está técnica desenvolvida no presente estudo seja uma nova opção complementar para a formação de viveiros matrizes de cana-de-açúcar com maior nível de sanidade.

Sugarcane leaf scald disease: growth in vitro of the causal agent (Xanthomonas albilineans), diagnosis by indicator plant and cure by thermotherapy in vivo and meristem tip culture
Abstract in English
The growth curve in vitro of Xanthomonas albilineans in modified Wilbrink medium was determined at 28°C and in the dark; its maximum stationary phase was reached with 7,5 to 8.5 x 108 CFU/ml, corresponding to an optic density from 0.15 to 0.20 in wave length of 625 nm, and 132 to 144 hours of incubation; the addition of 20% of dead cell extract of X. albilineans in modified Wilbrink medium, led to a reduction of the lag-phase period. However, the number of viable cells became smaller. By studying the behavior of X. albilineans in different temperature conditions, it was found that its growth in vitro at 40°C is negligible. However, the patogenicity is maintained up to 96 hours; the growth index is reduced, but the patogenicity is not lost when alternation is accomplished from 28°C to 40°C and is maintained up to 72 hours when the temperature is changed from 28°C to 40°C, but in this case the growth is not accomplished. By testing the Cuba sweet corn as an indicator plant for sugarcane leaf scald disease, it was verified that it shows symptoms when inoculated with stalk juice presenting vascular discolouration, but it is easier when the inoculum is from susceptible varieties, not presenting latent infection. In axenic culture, just a few bacterial cells inoculated by injection in foliar spindle of the Cuba sweet corn can cause leaf scald disease, which foliar symptoms are influenced by the conditions of luminosity and vascular discolouration by the presence of other microorganisms as Fusarium moniliforme and Erwinia herbicola. The Cuba sweet corn can, however, be used as an indicator plant in any growth phase, as a reliable and relatively feasible method for detection of leaf scald disease. The utilization of Cuba sweet corn led to a discovery of a possible natural method of bacterium dissemination through guttation droplets, since it shows symptoms when inoculated with droplets exeded from the interfecd sugarcane leaves, thus suggesting that leaf scald disease can be propagated in the field without man's interference. By testing the possibility of curing this disease, it was found that thermotherapy in vivo up to 40 days o at 40°C was not an efficient method. The association of thermotherapy in vivo and meristem tip culture led to a high percentage of cure. However, even with the application of gibberellic acid to obtain longer stalks and exposure of the plants to a continuous light, to increase their resistance, leaf scald disease was present in some explants tested. Since there is no natural cure for this disease in susceptible varieties and hot water treatment at 50.5°C/2 hours is also not efficient, it is suggested that the method developed in this study can be a new complementary option for the formation of matrix sugarcane nurseries with a higher level of sanity.
