O objetivo do projeto foi o desenvolvimento, descrição e otimização de protocolos de cultura de tecidos em melão amarelo (Cucumis melo var. inodorus Naudin), organogênese e embriogênese somática, que possam ser utilizados em trabalhos de transformação genética. O projeto consistiu na indução da organogênese in vitro, com a obtenção de gemas adventícias, e na indução da embriogênese somática, com obtenção de embriões somáticos pelo cultivo in vitro. A organogênese in vitro foi induzida pelo cultivo in vitro de discos de folha e segmentos de cotilédone, obtidos diretamente de sementes maduras. A embriogênese somática foi induzida pelo cultivo in vitro de segmentos de cotilédone. Os explantes foram inoculados no meio de cultura MS (Murashige & Skoog, 1962), suplementado com diferentes tipos e concentrações de reguladores vegetais, visando a obtenção de sistemas eficientes de regeneração de plantas. Para indução da organogênese in vitro, foram testados os reguladores vegetais: BAP (benzilaminopurina), 2-iP (2-isopenteniladenina), cinetina, TDZ (thidiazuron) e a combinação TDZ + NAA (ácido naftaleno acético). Para indução da embriogênese somática foram testados: IAA (ácido indolacético), NAA,2,4-D (ácido 2,4-diclorofenoxiacético), 2,4-D + NAA, 2,4-D + BAP e 2,4-D + TDZ. Para a indução de gemas adventícias, o regulador vegetal mais eficiente foi BAP, nas concentrações 0,5 a 1,0mg/L. Para indução de embriões somáticos, a combinação de 2,4-D + TDZ, nas concentrações de 2,5 a 7,5 mg/L e 0,1mg/L, respectivamente, mostrou maior eficiência no processo. Foram realizadas a análise morfológica e histológica, possibilitando a descrição dos dois processos morfogênicos. Na organogênese in vitro, cortes histológicos mostraram a formação de muitas protuberâncias e primórdios foliares, havendo a formação de poucas gemas adventícias. Na embriogênese somática, o estudo histológico mostrou a formação de muitos embriões somáticos mal formados, sem a presença de meristema caulinar, justificando a baixa conversão dos embriões somáticos

Tissue culture of yellow melon (Cucumis melo var. inodorus Naudin) : in vitro organogenesis and somatic embryogenesis

The objective of this project was the development, description and optimization of tissue culture protocols of yellow melon (Cucumis melo var. inodorus Naudin), including organogenesis and somatic embryogenesis, that can be used in studies of genetic transformation. The project consisted of the induction of organogenesis in vitro, with the formation of adventitious buds, and also the induction of somatic embryogenesis, with the formation of somatic embryos. In vitro organogenesis was induced by in vitro cultivation of leaf discs and cotyledon segments obtained from mature seeds. Somatic embryogenesis was induced by in vitro cultivation of cotyledon segments. The explants were inoculated in MS culture medium (Murashige & Skoog, 1962), supplemented with different types and concentrations of plant growth regulators, aiming to obtain efficient systems of plant regeneration. For in vitro organogenesis the following plant growth regulators were tested: BAP (benzylaminopurine), 2-iP (2-isopentenyladenine), kinetin, TOZ (thidiazuron) and a combination of TOZ + NAA (naphtaleneacetic acid). For somatic embryogenesis induction the following growth regulators were used: IAA (indolacetic acid), NAA, 2,4-0 (dichlorophenoxyacetic acid), 2,4-0 + NAA, 2,4-0 + BAP and 2,4-0 + TOZ. The induction of adventitious buds was more efficient with BAP at concentrations of 0.5 to 1.0 mg/L. For the induction of somatic embryos, the combination of 2,4-0 + TDZ at concentrations of 2.5 to 7.5 and 0.1 mg/L, respectively, showed the best efficiency. Morphological and histological analyses were done, allowing for the description of both morphogenic processes. In in vitro organogenesis histological sections showed the formation of many protuberances and leaf primordia, with few adventitious buds being formed. The histological study of somatic embryogenesis showed the formation of many malformed somatic embryos, lacking the shoot apical meristem, justifying the low conversion of the somatic embryos.
