Com o objetivo de encontrar uma alternativa metodológica visando a redução período de pré-condicionamento (tempo de embebição) que antecede o processo de coloração das sementes para a realização do teste de tetrazólio, foram estudados 4 períodos de embebição, 4h, 6h, 8h e 10h em 2 temperaturas (35°C e 42°C), tendo como testemunha o procedimento padrão de 16h de embebição a 25°C; em 15 lotes de sementes de soja fiscalizada dos cultivares Ocepar 4-Iguaçu, BR-16, IAS-5, BR-30 e BR-4. Foram realizados, também, testes de germinação, envelhecimento acelerado, condutividade elétrica e emergência das plântulas em areia, visando estabelecer correlação com os diferentes períodos de embebição/temperaturas utilizados para os 5 cultivares. Os estudos foram desenvolvidos nos laboratórios de sementes da Escola Superior de Agricultura "Luiz de Queiroz" - ESALQ, da Universidade de São Paulo - USP, em Piracicaba, SP, e do Centro Nacional de Pesquisa de Soja - CNPSo, da Empresa Brasileira de Pesquisa Agropecuária - EMBRAPA, em Londrina, PR. A análise dos dados e a interpretação dos resultados obtidos no presente trabalho, permitiram concluir que há possibilidade da redução do período de tempo necessário para a condução do teste de tetrazólio em amostras de sementes de soja, visando tanto a avaliação da germinação potencial (TZ 1-5) como a do vigor (TZ 1-3). A temperatura empregada para o pré-condicionamento das amostras é fundamental. Assim, a embebição das sementes a 42°C durante 8h a 10h e, eventualmente, durante 6h, permite o posterior desenvolvimento de coloração nítida e suficiente para exame adequado das áreas vitais das sementes e a identificação de sintomas de deterioração causados por "umidade", danos provocados por percevejos e ocorrência de injúrias mecânicas. De maneira geral, sementes dos cultivares em estudo apresentavam bom aspecto de coloração no teste de tetrazólio, quando o grau de umidade das sementes atingiu valor mínimo de 36%.

Alternative methodology for the tetrazolium test for soybean seeds

The objective of this study was to determine an alternative methodology that can reduce the preconditioning period (imbibition time) before the staining process in the tetrazolim test for soybean seeds. Fifteen lots of “fiscalized” soybean seeds of the cultivars OCEPAR 4-Iguaçu, BR-16, IAS-5, BR-30 and BR-4 with different levels of quality, were submitted to f our periods of imbibition (4, 6, 8 and 10 h) under two temperatures (35°C and 42°C). The standard procedure used for soybean seeds in the tetrazolium test (16 h/25°C) was used as the control. Additional tests were performed on these seeds: standard germination, accelerated aging, electric conductivity and emergence in sand. The results of these tests were correlated with the ones obtained by the tetrazolium test at the different preconditioning periods. The analyses were carried out in the seed laboratories of "Escola Superior de Agricultura Luiz de Queiroz" - ESALQ / University of São Paulo, Piracicaba, SP, and National Center for Soybean Research - EMBRAPA / CNPSo, Londrina, PR, Brazil. It was concluded that it is feasible to reduce the preconditioning period in the tetrazolium test for determining vigor [TZ (1-3)] and germination [(TZ (1-5)] potentials for soybean seeds. The temperature used during the preconditioning process of the seeds affects the precision of the test: preconditioning at 42°C for 8 to 10 h, and eventually for 6h, results in adequate staining patterns of the seed structures. This allows the identification of the typical patterns that result from seed deterioration (field weathering), stink bug damage and mechanical damage. In general, seeds of the studied cultivars presented adequate staining pattern by tetrazolium when seed moisture content reached the minimum value of 36%.
