O presente trabalho teve como objetivo a obtenção de biótipos estáveis de Trichoderma spp., resistentes ao fungicida benomyl, através do uso da irradiação ultravioleta, visando o controle integrado de Sclerotinia minor em plantas de alface (Lactuca sativa L.). Dos três isolados de T. harzianum e de T. viride expostos à luz U.V., o T. harzianum (TW5) foi o único do qual se obteve colônias resistentes a 2.000 ppm de benomyl. Os dez novos biótipos selecionados, após quatro repicagens sucessivas em meio de cultura, na ausência de benomyl, mantiveram as características de crescimento e esporulação quando colocados novamente para crescer em meio BDA com benomyl. Os novos biótipos apresentaram variações em relação ao crescimento, esporulação e morfologia das co1ônias; ao tamanho e freqüência de conídios uninucleados e/ou binucleados; ao comportamento em relação a esporulação e diâmetro de crescimento em meios de cultura com os fungicidas benomyl (2.000 ppm), thiabendazole (1.000 ppm), tiofanato de metila (1.000 ppm) e iprodione (1.000 ppm) ao antagonismo contra S. minor, S. sclerotiorum e S. rolfsii in itro, e aos padrões de eletroforese para esterase e proteínas totais. Em casa de vegetação, o efeito exercido pelo fungicida benomyl, associado ou não a três dos novos biótipos (2B1, 2B2 e 2B6) foi variável em relação ao controle de S. minor em alface. Os tratamentos com os novos biótipos 2B1 e 2B6 não foram eficazes no controle da doença. No entanto, o biótipo 2B1 associado ao benomyl, aumentou o nível de controle de S. minor, e o biótipo 2B6 foi menos eficiente do que quando associado ao benomyl. O biótipo 2B2 associado ou não ao benomyl, apresentou 100% de antagonismo ao S. minor.

Obtainment and characterization of new biotypes of Trichoderma harzianum, Rifai, resistant to benzimidazole fungicides through ultraviolet light

The objective of the present research was to obtain stable biotypes of Trichoderma spp, resistant to benomyl using ultraviolet light, for the integrated control of Sclerotinia minor in lettuce (Lactuca sativa L.). From the exposition of three isolates of T. harzianum and of T. viride to the U.V. light, T. harzianum (TW5) was the only to produce resistant colonies to 2.000 ppm of benomy. The ten new biotypes selected after four successives transplants in the absence of benomyl, they maintained the growth and sporulation characteristics when they were grown again in PDA medium with benomyl. The new biotypes presented variation in regard to growth, sporulation and morphology between their colonies; difference of size and frequence of unnucleated and/or binucleated spores; behavior in relation of sporulation and growth diameter in culture media containing benomyl (2.000 ppm); methyl thiophanate (1.000 ppm); thiabendazole (1.000 ppm) and iprodione (1.000 ppm); antagonism against S. minor, S. sclerotiorum and S. rolfsii "in vitro"; and the electrophoretic patterns proteins for esterases and total proteins. In greenhouse conditions, the effect benomyl associated or not with three biotypes (2Bl, 2B2 and 2B6) showed variation in relation to S. minor control in lettuce. The treatments of the new biotypes 2Bl and 2B6 were not efficient on the disease control. However, biotype 2Bl associated with benomyl increased the level the of S. minor control, and the biotype 2B6 was less efficient compared when it was associated or not with benomyl. The biotype 2B2 associated or not with benomyl, showed 100% of antagonism to S. minor.
