O presente trabalho foi desenvolvido com o objetivo de ampliar o conhecimento sobre a variabilidade genética natural existente em espécies do fungo Trichoderma e adicionar técnicas auxiliares para distinção de espécies nesse fungo. Nesse sentido. foram avaliadas 14 diferentes linhagens, quanto ao padrão eletroforético de isoenzimas, polimorfismo de tamanho de fragmentos de restrição (RFLP) e polimorfismo de DNA amplificado ao acaso (RAPD). Na técnica de eletroforese de isoenzimas, o sistema para esterases foi o que apresentou polimorfismo entre as linhagens. Na técnica de RFLP, duas sondas foram testadas: a sonda CBH1.1 que apresentou homologia somente com duas linhagens de Trichoderma e a sonda β-tubulina que apresentou homologia com todas as linhagens e o polimorfismo entre elas. Na técnica de RAPD dois "primers" foram utilizados e apresentaram resultados semelhantes. As linhagens puderam ser divididas em grupos de similaridades e diferenciações de linhagens da mesma espécie mas de locais diferentes puderam ser detectadas. As três técnicas utilizadas apresentaram resultados satisfatórios para análise de polimorfismo e podem ser utilizadas como técnicas complementares na identificação e classificação de espécies desse gênero.

Evaluation of genetic variability of Trichoderma by isozyme, RFLP and RAPD
Abstract in English
The present work was developed aiming to amplifie the knowledge about natural genetic variability existent in species of Trichoderma fungus and add complementary techniques to distinguish species. This way, 14 different strains were evaluated concerning isozyme electrophoretic patterns, restriction fragment length polymorfism (RFLP) and random amplified polymorphic DNA (RAPD). In the isozyme technique, esterase system was the one showing polymorphism among strains. In the RFLP technique two probes were tested. The probe CBHl.l showed homology only with two strains of Trichoderma. The probe β-tubulin showed homology with all strains and polymorphism among them. In the RAPD technique, two primers were tested and the results were similar. The strains were divided in similarity groups and differentations of strains of different places was observed. These techniques showed satisfying results and can be used as an alternative aid for polymorfism analysis in Trichoderma taxonomy.
