Este trabalho teve como objetivo a avaliação de diferentes metodologias para a obtenção de heterocários do fungo filamentoso Aspergillus niger, linhagens 10V10 e 391H6. Foram avaliadas a metodologia parassexual clássica, fusão química de protoplastos e a eletrofusão de protoplastos. Também foram avaliadas a capacidade de produção de ácido cítrico das linhagens como também a atividade celulolítica. Primeiramente foram obtidos mutantes auxotróficos e morfológicos por indução através de luz ultravioleta. Procedeu-se então o cruzamento pela via parassexual clássica, por fusão de protoplastos com o auxílio de polietileno glicol e por eletrofusão. A atividade celulolítica deu-se através da digestão de papel de filtro por enzimas produzidas pelo fungo. A produção de ácido cítrico foi avaliada por titulação do meio de fermentação. Os resultados demostraram que a eletrofusão de protoplastos foi a metodologia mais conveniente para a obtenção de hetrocários; as linhagens apresentaram uma alta atividade celulolítica e pelo menos a linhagem 10V10 demostrou produzir ácido cítrico.

Parasexuality and electrofusion in Aspergillus niger
Abstract in English
Different methodologies to obtain heterocaries of Aspergillus niger were evaluated. ln this work were used two industrial citric acid producer strains: 10V10 and 391H6. The classical parassexual methodology, chemical fusion of protoplasts and electrofusion of protoplasts were analyzed. It was also evaluated the ability of citric acid production and cellulolytic activity. First of all, auxotrofic and morphological mutants were obtained by induction with ultraviolet light. After these crosses via parassexual cycle, protoplast fusion with polyetilen glycol electrofusion were done. The cellulolytic activity was done by the digestion of filter paper by the enzymes produced by the fungi. The citric acid production, by its turn, was evaluated by title of the fermentation media. The results show that. the electrofusion of protoplasts was the most suitable methodology to obtain heterocaries; both strains show a high cellulolytic activity and, not unsurprisingly, the strain 10V10 showed citric acid production.
