A utilização de linhagem de Aspergillius nidulars para a detecção de substâncias que acarretam mutação de ponto, tem recebido relativa atenção por parte dos pesquisadores. Com aquela finalidade, foi verificada na presente pesquisa a possibilidade de utilização de uma linhagem (CHL-13) que é resistente e parcialmente dependente do cloroneb para detecção de mutagênicos físicos. Verificou-se a possibilidade dessa linhagem ser utilizada para tal mister. Foram, estatisticamente, selecionadas para análise genética seis linhagens resistentes e duas sensíveis, das 201 obtidas através do método de emersão de setores. Pelo teste de alelismo verificou-se que os fatores genéticos pertenciam a um mesmo locus testes de heterocário mostraram que fatores extranucleares não influíram na manifestação do caráter resistência ou sensibilidade ao cloroneb; por análise mitótica concluiu-se que o locus em epígrafe estava no grupo de ligação III; análise meiótica indicou que o locus para resistência ou sensibilidade ao cloroneb situava-se a aproximadamente 30 unidades de recombinação do locus galA1, além de ter sido verificado através de oito diferentes produtos de haploidização, que genes de outros loci não influíram na sensibilidade da linhagem ao cloroneb

The utilization of Aspergillius nidulars strains for the detection of substances that produce point mutation, have received little attention. This study verifies the possibility of using a strain which is resistant and partially dependent on chloroneb. Six resistant and two sensitive strains were selected statistically for genetic analysis. From there 201 sectors were obtained using the method of sector emersion. These presented perfect delineation, emerged from colonies of strain CHL-13 and were obtained by radiating microcolonies with gama ray and ultra-violet radiation. Allelism tests have shown that only one locus was involved; the heterokaryon test proved that extranuclear factors were not influencing the resistance or sensitivity of the strain to showed that the locus was in chloroneb; mitotic analyses linkage group III; meiotic analyses indicated that the locus for resistance or sensitivity to chloroneb was situated approximately 30 recombination units from locus galA1; and finally it was verified after using eight different post-haploidie sensitive products, that genes at other loci did not influence the sensitivity of the strain to chloroneb
