A espécie Fusarium moniliforme é encontrada freqüentemente em sementes de milho, sendo essa possivelmente a sua forma de inóculo no campo. Fusarium spp. são associados com plantas sintomáticas e assintomáticas. Sendo que esse fungo pode ser tanto um agente causal primário de doença, um invasor secundário ou um endófito. Neste presente trabalho, procedeu-se ao isolamento de fungos endofíticos de sementes de duas populações de Zea mays (BR105 e BR106). Dentro das diferentes linhagens de milho ensaiadas, encontrou-se Fusarium em uma freqüência que variou de 0 a 100% em relação ao total de fungos isolados. A análise molecular dos isolados foi realizada utilizando-se a técnica de RAPD. Foram analisadas 21 linhagens de F. moniliforme usando-se 10 iniciadores aleatórios da Operon Technologies (Kit X). Observou-se uma variabilidade genética entre os isolados endofíticos de F. moniliforme, analisados por RAPD. Esses isolados puderam ser agrupados por local de isolamento. Também foi observada uma relação entre os isolados, as linhagens de sementes e a população de milho ensaiada. Essa relação ocorreu de forma parcial, tendo sido mais evidente nos isolados de um local do que no outro. Objetivando-se estudar a base molecular da patogenicidade e especialização ao hospedeiro em isolados endofíticos de F. moniliforme, desenvolveu-se um sistema de transformação genética baseado no gene da nitrato redutase de Fusarium oxysporum. Foram obtidas freqüências de transformação que variaram de 30 a 60 transformantes por μg de DNA. Objetivando-se ainda um futuro monitoramento de linhagens endofíticas em milho, bem como uma utilização das mesmas como portadores de genes para a transformação genética de plantas, obteve-se linhagens marcadas com o gene da enzima GUS. A observação da expressão do gene em questão em um transformante no interior da raiz de milho, foi realizada com sucesso. Neste trabalho também foi observada a transposição genética do elemento Impala de F. oxysporum em F. moniliforme.

The Fusarium moniliforme specie is often found in seeds of mayze and this it is the possible way of inoculum in field. Fusarium spp. are associated with sintomatics and assintomatics plants. This fungi can be found as a primary causal agent of disease, a secundary invader or an endophyte. In the present work, endophytic fungi was isolated from two populations of Zea mays (BR 105 and BR 106). ln differents strains of maize assessed, Fusarium have been found in a range frequency of 0 to 100% in relation with the total isolated fungi. The molecular analysis of isolates was performed using RAPD technique. Twenty-one F. moniliforme strains were analyzed using ten randomic primers Operon Technologies (Kit X). A genetic variability between endophytics strains have been observed by RAPD. Such strains were clustered by local origin. The association between strains, seeds lines and maize population, also were possible. The clustering was partial, and were more evident in strains from on place than another. With the aim of to study the molecular basis of pathogenicity and specialization of F. monilifonne endophitic strains to host, was developed a system of genetic transformation based on nitrate reductase gene of Fusarium oxysporum. Transformation frequencies of 30 to 60 transformants per μg DNA was acquired. With the aim of a future monitoration of endophytic strains in mayze, as well as his use as a vector of genetic transformations of plants, marked strains were obtained with the GUS gene. The observation of gene expression in a transformant into mayze roots, was made successfully. ln that work, the genetic transposition of Impala element of F. oxysporum in F. moniliforme was also observed.
