A seringueira, apesar de sua importância pela produção de borracha natural, é pouco estudada, especialmente em aspectos básicos como citogenética e biologia reprodutiva e isto deve-se, em parte, à dificuldade em se trabalhar com tal espécie arbórea. Porte alto, longos períodos de imaturidade e ciclo biológico, baixa produção de frutos, sementes recalcitrantes, são alguns dos fatores limitantes. O presente estudo teve como objetivo investigar aspectos citogenéticos ( caracterização de cromossomos . somáticos e análise da macrosporogênese ), morfológicos ( estudo da morfologia floral e taxa de hermafroditismo) e fisiológicos ( enraizamento de plantas jovens em meio líquido e de ápices caulinares in vitro ), em Hevea brasiliensis. Três clones foram estudados, RRIM 527, RRIM 600 e GT l (macho-estéril). Os cromossomos somáticos foram avaliados por técnicas citológicas convencionais e bandamento C. Foram detectadas algumas regiões heterocromáticas. Através de metodologia de clareamento de óvulos usando salicilato de metila, observou-se que a macrosporogênese é normal, nos três clones, culminando com a formação de sacos embrionários octanucleados. A técnica permitiu a observação de grande número de óvulos e nenhuma irregularidade meiótica foi detectada. As alterações observadas ( como exceções) foram morfológicas. Botões florais com morfologia externa de femininos foram analisados sob microscopia eletrônica de varredura quanto à sua estrutura. Os resultados mostraram que do total de flores avaliadas, 71,49% eram hermafroditas e 28,51 % exclusivamente femininas. As flores hermafroditas possuem ovário normal em cuja base as anteras dispõem-se, circundando-o completa ou parcialmente. Tais anteras são geralmente menores que as normais, mas em grande parte funcionais e com grãos de pólen. Do total de flores hermafroditas, 29,83% apresentou anteras residuais e 71,17% estames desenvolvidos. Ensaios de enraizamento de plantas de seis e 18 meses de idade foram realizados em quatro tipos de meio líquido (solução de Sarruge e água, com e sem fornecimento de NAA). Os resultados mostraram que água, com borbulhamento contínuo de ar, foi o melhor meio e grande quantidade de raízes foi obtida nos dois tipos de plantas. Fornecimento de NAA, mesmo em baixas dosagens, inibiu o enraizamento. Ápices caulinares de diferentes origens foram utilizados em ensaios de enraizamento in vitro mas somente aqueles obtidos de plântulas recém germinadas mostraram bons resultados. Foi determinado um protocolo de estabelecimento e enraizamento usando-se este tipo de material como explante.

Rubber tree, in spite of your importance due to production of natural rubber is little studied, specially in basic aspects like cytogenetics and reproductive biology and this is partially due to the dificulty in working with this species. Height, long imaturity period and biological cicle, low fruit set, recalcitrant seeds are some of the limitant factors. This study was aimed to investigate cytogenetics (somatic chromosomes characterization and megasporogenesis analysis ), morphological (floral morphology and hermaphroditism rates) and physiological (rooting of young plants in liquid medium and of shoots in vitro) aspects in Hevea brasiliensis. Three clones were studied, RRIM 527, RRIM 600 and GT 1 (male-sterile ). Somatic chromosomes were avaluated by conventional cytological techniques and C-banding. Some heterochromatic regions were detected. By ovule clearing technique using methyl salycilate it was observed that megasporogenesis is normal in the three clones, culminating with the formation of octanucleated embryonary sacs. This technique allow the analysis of a great number of ovules and no irregularities were detected. Toe observed alterations ( exceptions) were morphological. Flower buds with externai morphology of female were analysed by scanning electron microscopy with regard of its structure. Results showed that 71,49% of total flowers were hermaphrodites and just 28,51 % exclusively females. Toe hermaphrodite flower has a normal ovary surrounded by stamens. This anthers are generally smaller than those of male flowers, but are functional with polen grains. Of the total number of hermaphrodite flowers 29,83% had residual anthers, not completely developped, but in the great part of the analysed hermaphrodite flowers (70,17%) stamens were developped. Rooting tests of six and 18 months aged plants were realized in four types of liquid medium (Sarruge's solution, water, with or no NAA). Results showed that water was the best medium and a great number of rootswas obtained in the two types of plants. Supplying NAA inhibit rooting even in lower dosage. Shoots from differents origins were utilized on in vitro rooting tests but just that from plantlets showed positive results. It was determinated a protocol of in vitro establishment and rooting for this material.
