mancha foliar provocada por Septoria tritici Rob. é uma das principais doenças do trigo em mais de 50 países, sendo responsável por significativas reduções na produtividade desta cultura. Os objetivos do presente trabalho foram: (i) Estudar a interação resistência x estádio fenológico no patosistema trigo - Septoria tritici em 77 progênies F11 de trigo derivadas do cruzamento triplo Tadinia x (Yecora rojo x UC554); (ii) Determinar se existe especialização fisiológica em isolados de Septoria tritici originários de Argentina e EUA através da análise de sua patogenicidade em cultivares e linhagens de trigo (Triticum aestivum; (iii) Avaliar a resistência a Septoria tritici em progênies F11 de trigo derivadas do cruzamento Tadinia x (Yecora rojo x UC554) em diferentes estádios fenológicos visando posterior análise com marcadores moleculares e (iv) Detectar genes de resistência a Septoria tritici através de marcadores microssatélites e AFLPs nestas progênies. A técnica de mapeamento empregada foi a do Bulked Segregant Analysis, ou análise da mistura de DNA de indivíduos segregantes. Os agrupamentos (extremos) foram constituídos com base em avaliações da resistência realizadas em casa de vegetação e campo, envolvendo três estádios fenológicos: plântula, perfilhamento e folha bandeira e dois anos agrícolas (1999 - 2000). Os extremos foram analisados com marcadores microssatélites e AFLPs. Foram feitos testes com 307 primers de microssatélites pertencentes às séries wms e wmc, selecionando 262 primers polimórficos. Na análise com marcadores AFLPs, foram utilizadas combinações de dois primers seletivos Pst I com dez primers Msel. Foram detectadas diferenças altamente significativas (p ≤ 0,0001) entre progênies para área foliar lesionada (AFL) em todos os estádios fenológicos estudados. Análises de variância conjuntas para AFL indicaram uma interação significativa entre progênies e estádios fenológicos (p ≤ 0,0001), indicando a necessidade de se avaliar a resistência em mais de um estádio fenológico para garantir correta seleção de genótipos resistentes. Foram detectadas variações na agressividade entre isolados de Septoria tritici e especialização fisiológica dos mesmos, sugerindo a necessidade de testes de patogenicidade como uma prioridade na seleção de materiais de trigo resistentes a este patógeno, já que a agressividade de cada isolado pode variar significativamente de acordo com o genótipo do hospedeiro, acarretando problemas na avaliação e seleção de genótipos resistentes. Não foi possível detectar genes de resistência na população Tadinia x (Yecora rojo x UC554). Três fatores podem ser apontados como responsáveis por este insucesso: 1) população de mapeamento derivada de cruzamento triplo; 2) presença de progênies não pertencentes ao cruzamento (fora do tipo) e 3) comportamento não esperado do parental UC554 em relação à resistência a Septoria tritici.

Detection of resistance genes to Septoria tritici Rob. in wheat [Tritieum aestivum (L.)]

Leaf blotch caused by Septoria tritici Rob. is one of the most important diseases of wheat in more than 50 countries, being responsible for significant yield losses. The objectives of the present work were: (i) To study the interaction between resistance x phenological stage in the pathosystem wheat - Septoria tritici in 77 F11 progenies of wheat derived from the three-way cross Tadinia x (Yecora rojo x UC554); (ii) To Determine the degree of physiologic specialization among Argentinean and USA isolates of Septoria tritici through inoculation on cultivars and lines of bread wheat (Triticum aestivum L.); (iii) To evaluate the resistance to Septoria tritici in F11 wheat progenies derived from the three-way cross Tadinia x (Yecora rojo x UC554) in different phenological stages seeking the subsequent analysis with molecular markers, and (iv) To detect resistance genes to Septoria tritici through microsatellites and AFLP markers. The mapping technique used was Bulked Segregant Analysis, or analysis of mixtures of DNA of segregant individuals. Bulk extremes were composed based on greenhouse and field disease assessments made in three phenological stages: seedling, tillering and flag leaf, and two years (1999-2000). Resistant and susceptible extremes were analyzed with microsatellites and AFLP markers. 307 microsatellite primers belonging to the series wms and wmc were used, from which 262 polymorphic ones were chosen for further analysis. In the analysis with AFLP markers, combinations of two Pstl with ten Msel selective primers were used. Highly significant differences for damaged leaf area were found (p ≤ 0.0001) among progenies in all phenological stages studied. Joint analyses of variance for DLA indicated a highly significant interaction between progenies and phenological stages (p ≤ 0.0001), indicating the need to evaluate resistance in more than one phenological stage to guarantee the correct selection of resistant genotypes. Variations in aggressiveness among strains of Septoria tritici were detected, indicating the need of pathogenicity tests as a priority in the selection of resistant wheat materials, since the aggressiveness of each isolate can vary significantly depending on the host genotype. It was not possible to detect resistance genes in the population Tadinia x (Yecora rojo x UC554). Three factors can be pointed as responsible for this failure 1) Employment of a three way cross mapping population; 2) Presence of out-of- type progenies, and 3) Unexpected behavior of UC554 when inoculated with Septoria tritici.
