O interesse científico pelo gênero Stylosanthes tem aumentado desde a última década devido a descobertas de muitas espécies que são potencialmente importantes como forrageiras para áreas tropicais. Para melhorar estas espécies de interesse agronômico é necessário conhecer e classificar adequadamente o material em estudo. Entretanto, a classificação taxonômica do gênero é controvertida, especialmente dos taxons relacionados com Stylosanthes guianensis: a literatura tem considerado este grupo de plantas ora como variedades botânicas de S. guianensis, ora como espécies distintas. O objetivo do presente trabalho foi estudar o gênero sob o ponto de vista citotaxonômico: nove espécies diplóides nativas do Brasil (seis relacionadas com o "complexo guianensis") e quatro variedades botânicas de S.guianensis, descritas pela EPAMIG, foram examinadas. Empregou-se a metodologia utilizada para o exame de cromossomos somáticos de plantas, com um pre-tratamento das pontas de raiz com 8-hidroxiquinolina 0,03%, seguido da coloração pelo método de Feulgen. Empregou-se ainda a metodologia de bandamento-C que revela a presença de blocos heterocromâticos nos cromossomos metafásicos. Para comparar os taxons, foram utilizados os dados de relação de braços, comprimento relativo, Índice de simetria (TF) além do comprimento do lote haplóide. Estes dados foram submetidos a dois tipos de tratamento estatístico: uma análise univariada e uma análise multidimensional. Os resultados obtidos indicaram que o gênero Stylosanthes apresenta um cariótipo padrão para todas as espécies analisadas com 2n = 20, porém foi possível detectar pequenas diferenças que caracterizam cada material. Entre os taxons pertencentes ao "complexo guianensis" foi possível discriminar cada uma das quatro variedades botânicas de S.guianensis (S.g.vulgaris, S.g.microcephala, S.g.canescens e S.g.pauciflora) e as espécies S.gracilis e S. campestris. Ainda dentro do complexo, os cariótipos de S.g.vulgaris e de S. pilosa mostraram-se semelhantes ao de S.grandifolia e o cariótipo de S.montevidensis ao de S.gracilis. Foi possível ainda caracterizar citologicamente as três espécies não pertencentes ao complexo, S.humilis, S. macrocephala e S.viscosa que mostraram cariótipos bem distintos. Os materiais analisados não mostraram blocos heterocromâticos evidentes quando submetidos ao tratamento de bandamento-C. Os resultados do presente trabalho sugerem que os mecanismos relacionados com o processo de especiação em Stylosanthes envolvem basicamente DNAs eucromâticos, sobretudo com alterações do tipo inversão pericêntrica. Além disso, sugere-se que pequenas alterações no conteúdo de DNA eucromâtico possam ter ocorrido no curso de evolução do gênero.

Cytotaxonomic study of brazilian species of the genus Stylosanthes Sw

The scientific interest in the genus Stylosanthes Sw. has been increased over the last decade by discoveries of many species that are potentially important for forage tropical areas. In order to improve these species of agronomic interest, it is necessary to know and correctly classify the plant material being studied. However, the classification of the genus is unresolved, specially in the taxa related to Stylosanthes the literature has considered this group of plants either as guianensis: botanical varieties of S. guianensis or as different species. The aim of the present work was to study the genus on the citotaxonomic point of view: nine diploid species from Brazil (six of them related to the “guianensis alliance”) and four botanical varieties of S. guianensis described by EPAMIG, were examined. The standard methodology was used to analyse the plant somatic chromosomes with a pre-treatment of the root tips using 8-hydroxiquinolin 0,03%,followed by the Feulgen staining method. The C-banding methodology which reveals be presence of heterochromatic blocks in the metaphase chromosomes was used as well. The data related to arm ratio, relative lenght, symetry index (TF) and haploid set lenght were used to compare the taxa by two kinds of statistical treatment: univariate analysis of variance and multivariate analysis. The results obtained indicated that the genus Stylosanthesshows a standard karyotype, wi th 2n = 20; nevertheless, differences in the morphology of the chromosomes were observed and each material was characterized. Among the taxa belonging to the “guianensis alliance” it was possible to discriminate each variety of Stylosanthes guianensis (S.g.vulgaris, S.g.microcephala, S.g.canescens and S.g.pauciffora) and the species S.gracilis and S.campestris. The karyotypes of S.g.vulgaris and S.pilosa were similar to that of S.grandifolia and the karyotype of S.montevidensis was similar to that of S.gracilis. Moreover, it was possible to characterize the three species not belonging to the “guianensis alliance”, S. humilis, S.macrocephala and S.viscosa, which showed differents karyotypes. Virtuals heterochromatic blocks were not observed in any of the species analysed after C-banding treatment. The results of the present work suggest that the chromosomal mechanisms related to the speciation process in Stylosanthes involve basically euchromatic DNAs, mainly with alterations of the pericentric inversion type. Besides, it has been suggested that small alterations in the euchromatic DNA content may have occurred in the evolution process of the genus.
