Neste trabalho, foram analisadas 25 populações de S. humilis H.B.K. cujas sementes foram coletadas em duas transeções (Norte-Sul e Leste-Oeste), ao longo de três regiões ecogeográficas do Estado de Pernambuco. Todas as famílias coletadas foram analisadas em laboratório através da técnica de eletroforese. Foram determinados e estudados 20 locos isoenzimáticos sendo 15 polimórficos, pertencentes a cinco sistemas: catalase, esterase, leucina aminopeptidase, malato desidrogenase e peroxidase. Foram analisadas em média também 20 progênies por família para determinar a taxa de cruzamento em algumas populações naturais. Com o objetivo de se avaliar a relação dos padrões de variabilidade aloenzimática obtida em laboratório e a influência da seleção natural nas diferentes condições ecogeográficas de origem de cada população, foram instalados dois ensaios no campo experimental de Anhembi, Piracicaba, sendo um para quantificar e analisar o desempenho dos caracteres morfológicos e agronômicos e outro para determinar a taxa de cruzamento usando locos marcadores (E1 e E2) homozigotos. Progênies do ensaio instalado para obtenção da taxa de cruzamento e das plantas mais e menos produtivas do outro ensaio também foram analisadas por eletroforese para a obtenção dos resultados que são discutidos neste trabalho. As conclusões básicas deste trabalho sao as seguintes: 1. Em S. humilis, o polimorfismo isoenzimático e uma função da variação ambiental. 2. A diversidade gênica (H), que é a melhor expressao da variabilidade genética é uma evidência da resposta imediata às condições ambientais principalmente clima, solo e tamanho da população. 3. A região do Agreste, apesar de ser a menor região estudada e suas populações ocorrerem a beira de rodovias e serem relativamente próximas, além de ser a região mais variável e de clima instável, comporta populações de maior variabilidade em todos os níveis genéticos estudados. 4. As duas regiões ecogeográficas extremas, Mata e Sertão apresentam padrões de variabilidade genética uniformes porém distintos, com menores valores na região da Mata, as quais são interligadas pelo padrão do Agreste que além de ser intermediário possui populações com valores superiores as outras duas. 5. Com base na estrutura genética das populações da região da Mata, concluiu-se que estas apresentam uma distribuição marginal, cujo estreitamento da variabilidade deve ter sido ocasionado pelo “efeito do fundador”, levando à deriva genética. 6. As populações do Agreste e Sertão devem pertencer ao centro de diversidade da espécie. 7. A curva de distribuição da frequência da taxa de cruzamento em ensaio de campo, acompanha a curva da frequência do florescimento. 8. Quanto maior a taxa de cruzamento maior e a variabilidade observada nas populações naturais e menor e a divergência genética. 9. Os resultados de três metodologias para medir a taxa de cruzamento foram equivalentes, com variações médias entre populações e regiões de 21% a 32%.10. As regiões climáticas não influem significativamente sobre a produção forrageira e sim sobre os caracteres relacionados à superficie foliar e comprimento do pecíolo foliar e internódio.11. A produção forrageira em S. humilis em ambientes heterogêneos é favorecida pela variabilidade genética.12. As populações provenientes de regiões de precipitação tardia apresentam maior produção de biomassa. 13. A variabilidade genética expressa pela proporçao de locos polimórficos e pela diversidade é inversamente proporcional à precipitação pluviométrica anual, o que faz esta espécie ser bem adaptada ao semi-árido. 14. A diversidade gênica expresssa pela quantidade de informação genética diferente (alelos diferentes) e pela heterozigosidade é positivamente correlacionada com a produtividade forrageira do S. humilis.

Genetic structure of populations of Stylosanthes humilis H.B.K. (Leguminosae) from three ecogeographic regions of the state of Pernambuco

Twenty five populations of S. humilis H.B.K. collected along two transects (North-South, East-West) in three ecogeographic regions of the State of Pernambuco, Brazil, were analysed at the level of families, through the technique of gel eletrophoresis. Twenty isoenzimatic loci of wich fifteen were polymorphic were established and studied. They belong to five systems: catalase, esterase, leucine aminopeptidase, malate dehydrogenase and peroxidase. Twenty progênies per Family were algo analysed to determine the crossing rate in natural populations. With the aim of evaluating the relatioship between the alloenzimatic patterns established in laboratory and the influence of natural selection in the regions of origin of each population, two field trials were conducted at the Experimental Station of Anhembi, Piracicaba, in order to quantify and to analyse the performance of morphological and agronomic characters, and to determine the crossing rate using homozygous marker loci (E1 and E2). Progenies from the trial conducted to obtain the crossing rate and the plants more or less productive from the other trial, were also analysed through electrophoresis to obtain the results discussed in this thesis. The basic conclusions are the following: 1. In S. humilis the isoenzimatic polymorphism is a function of the environmental variation. 2. The genetic diversity (H), whoch is the best expression of genetic variability, is evidence of the direct response to the environmental conditions specially climate, soil and populations size. 3. The populations from the Agreste region showed the largest variation at all levels studied, although this region was the smallest region studied and its populations occur along roadsides and are very close one to another. Besides this, this region is the most variable and unpretictable of all regions studies. 4. The two extreme ecogeographic regions, Mata and Sertão exhibit a pattern of genetic variability uniform but distinct, with the smallest values for the Mata region.The two regions are connected by the Agreste pattern, which is intermediate and shows populations with values greater than the other two regions. 5. Based on the genetic structure of the populations from the Mata region, it was concluded that these populations exhibit a marginal distribution, with a reduction in the variability due probably to founder effect which led togenetic drift. 6. The populations from the Agreste and Sertâo regions probably belong to the center of diversity of the species. 7. The curve of frequency distribution of crossing rate inthe field trials follows the curves of frequency of flowering. 8. The larger the crossing rate, the larger the variability observed in natural populations, and the smaller the genetic divergence. 9. The results from the three methodologies used to estimate the crossing rate were similar, ranging among populations and regions from 21% to 32%. 10. The climatic regions do not influence the forage production significantly, but influence leaf area and length of leaf and internode. 11. Forage production in S. humilis environments is favoured in heterogeneous by genetic variability.12. The populations from regions of late rains exhibit large forage production.13. The genetic variability expressed by the proportion of polymorphic loci and diversity is inversely proportional to the annual rain fall.14. The genetic diversity expressed by the quantity of different genetic information (different alleles) and by heterozygosity is positively correlated with forage production in S. humilis.
