Foram estudadas 19 estirpes de Rhizobium phaseoli quanto as suas características culturais, crescimento em meio definido e em meio definido e em meio com peptona e glicose. Todas as estirpes cresceram nesses meios, variando, entretanto, a intensidade do crescimento. Análises eletroforéticas em gel de agarose evidenciaram a presença de, pelo menos 1 plasmídio, na maioria das estirpes. Tais plasmídios foram tentativamente correlacionados com a produção de bacteriocina, com a nodulação e com a capacidade de fixar nitrogênio. Por eletroforese em gel de acrilamida foram determinados os padrões de esterases das estirpes, os quais parecem estar correlacionados com o seu local de origem. O crescimento em meio definido e a presença de plasmídios na maioria das estirpes ensaiadas abre possibilidades para estudos genéticos nas mesmas. Por outro lado, a verificação de que ê possível separar estirpes segundo seu padrão de bandas no que se refere a esterases vai permitir a utilização dessas marcas em estudos de inoculação de estirpes em campo.

Caracterization and genetical aspects of strains of Rhizobium phaseoli

Abstract in English
Nineteen strains of Rhizobium phaseoli were studied in relation to their growth characteristics including growth on defined and peptone-glucose medium. All tested strains were able to grow on these media. However the rates of growth varied according to the strains. Agarose gel electrophoresis have shown that most of the strains presented one or more plasmids. Such plasmids were tentatively correlated with bacteriocin production, nodulation and nitrogen fixation ability. Acrilamide gel electrophoresis have shown that several esterase patterns occurred in different strains and that these patterns can be related to the region from which such strains were isolated. Growth ability on defined medium and the presence of plasmids in the majority of the strains are features which can be used for future genetic studies in R. phaseoli. Also the fact that it was possible to group strains according to their esterases pattern opens the possibility for the use of such markers in inoculation studies on field experiments.
