O presente trabalho foi desenvolvido com a finalidade de se analisar genética e citologicamente a linhagem deteriorada V8 de Aspergillus nidulans (Eidam) Winter e os dissômicos por ela produzidos por não-disjunção mitótica. As estimativas das frequências de aneuplóides foram feitas através de semeaduras de conídios da linhagem V8 em meios completos com e sem desoxicolato de sódio. Paralelamente, foram estudados os possíveis agentes modificadores da frequência de aneuplóides: temperatura, pH e idade do conídio. Estudos citológicos foram conduzidos para comparar os tamanhos dos núcleos e das células de conídios das linhagens V8, dissômica e MSE, e analisar hifas ascógenas provenientes de cruzamentos MSE x dissômico. Os resultados obtidos permitiram chegar às seguintes conclusões: 1. Em virtude da baixa taxa de crescimento da linhagem V8, tornou-se desnecessária a adição de desoxicolato de sódio para melhor visualização dos aneuplóides. 2. O tipo de aberração cromossômica e a constituição genética da linhagem são fatores que influíram na frequência de aneuplóides. O tipo dissômico obtido e sua alta estabilidade reforçam a possibilidade do gene V8 aumentar a taxa de não-disfunção mitótica. 3. Dentro dos limites utilizados não houve efeitos da temperatura ou do pH na frequência de aneuplóides. A idade do conídio influiu na não-disjunção mitótica: conídios mais velhos produzem maior frequência de aneuplóides que os mais novos. 4. A análise genética do cruzamento Dissômico x MSE mostrou que o cromossomo adicional é transferido durante o processo meiótico. 5. Citologicamente, a linhagem V8 apresenta o diâmetro médio de seus núcleos maior que o da linhagem MSE e menor que o da linhagem dissômica. 6. A técnica citológica com melhores resultados para a coloração de núcleos de conídios e de hifas ascógenas das linhagens estudadas foi a HCl-Giemsa. A diluição do corante e o tempo de coloração e hidrólise foram adaptados para cada linhagem.

The present study was carried out to analyse genetically and cytologically a deteriorated V8 strain of Aspergillus nidulans (Eidam) Winter, and the dissomics produced by mitotic non-disjunction. Estimates of the frequency of aneuploids were made by plating out conidia of the VB strain on complete medium with and without sodium deoxycholate. In parallel, studies were made of factors which might influence the frequency of aneuploids: temperature, pH and the age of the conidia. Cytological studies were carried out to compare the sizes of the nuclei and cells of conidia of the V8, dissomic and MSE strains, and to analyse ascogenous hyphae produced by MSE x dissomic crosses. From the results obtained, the following conclusions can be drawn: 1) because of the low growth rate of the V8. Strain, it was not necessary to add sodium deoxycholate in order to distinguish aneuploids. 2. the type of chromosomal aberration and the genetic constitution of the strain are factors which influence the frequency of aneuploids. The type of dissomics obtained, and their high stability, suggest that the V8 gene increases the rate of mitotic non-disjunction. 3. within the limits tested, temperature and pH had na effect on aneuploid frequency, but age of conidia did have an effect: old conidia produce a higher frequency of aneuploids than younger ones. 4. genetic analysis of the dissomic x MSE cross showed that the addltional chromosome was not lost during meiosis. 5. Cytologically, the V8 strain shows a greater mean nuclear diameter than the MSE and a smaller one that the dissomic strain. 6. The cytological technique which gave best results for staining the nuclei of conidia and ascogenous hyphae of the studied strains was HCl-Giemsa. The dilution of the stain and the times of staining and hydrolysis were adjusted for each strain.
