No presente trabalho foram realizados estudos genéticos em Trichoderma pseudokoningii e verificada a atividade celulolítica da linhagem selvagem, mutante e recombinantes obtidos de heterocários. Com o objetivo de se obter mutantes auxotrófico, testou-se dois agentes mutagênicos, a luz ultravioleta e o ácido nitroso. Através de irradiação com luz ultravioleta foram obtidos dezenove mutantes autotróficos simples e dezoito com dupla deficiência, e dois mutantes morfológicos com ácido nitroso. Sete mutantes com dupla marcas autotróficas e morfológicas foram utilizados para a obtenção de heterocários. Colônias recombinantes foram obtidas diretamente dos heterocários em diferentes meios seletivo, sugerindo a ocorrência de parameiose nesta espécie. Colônias protróficas crescidas em MM eram binucleadas e de coloração verde, indicando um sistema não autônomo para a pigmentação de conídios. Estas colônias dicarióticas formaram setores em MC acrescido de benomyl, sugerindo a ocorrência de uma fase diplóide altamente instável e parassexualidade. Foi descrito um método para a obtenção e fusão de protoplastos. Colônias recombinantes foram recuperadas diretamente de produtos de fusão, numa freqüência aproximadamente 1000 vezes superior em relação às obtidas à partir de heterocário. Quanto a atividade celulolítica da linhagem selvagem e mutantes, observou-se que os valores de índices enzimáticos foram diretamente proporcionais ao maior ou menor grau de compactação das colônias. Colônias de linhagem selvagem originadas de protoplastos regenerados apresentaram índices de atividade celulolítica inferiores em relação às colônias originadas de conídios, indicando um efeito negativo da protoplastização sobre a atividade enzimática

In this work genetic studies were carried out in Trichoderma pseudokoningii in order to test the cellulolitic activity in wild and mutant strains and recombinants obtained from heterokaryons. Two mutagenic agents were tested aiming to obtain auxotrophic mutants-Utraviolet light and nitrous acid. Through UV light were obtained 19 simple auxotrophic mutants and 18 double mutants and through nitrous acid two morphological mutants. Seven color mutants carrying two auxotrophic markers were used to get heterokaryons. Recombinant colonies were directly recovered from heterokaryons onto different seletive media, indicating the occurrence of parameiosis. Prototrophic colonies grown in minimal medium were binucleated and green coloured, revealing a non autonomous system for conidia pigmentation. These dikaryotic colonies gave rise to sectors in complete medium plus benomyl suggesting the occurrence of a highly unstable diploid phase and parassexuality. It was described as well a method for protoplasts obtention and fusion. Recombinant colonies were directly recovered from fusion products in a frequency almost 1000 times higher than the recombinants from heterokaryons. For cellulolitic activity in wild and mutant strains it was shown that the enzymatic indexes were directly proportional to the higher or smaller degree of colony compactation. Colonies regenerated from protoplasts of the wild strain showed worst cellulolitic activity than colonies rosen from conidia. This is a negative effect of protoplastization on the cellulolitic activity. Electrophoretic patterns for esterase of the wild and mutant strains showed different profiles according to number and band position. It might be suggested a relation between band patterns and conidia coloration.
