Com o objetivo de estudar relações entre as atividades das enzimas fosfoenolpiruvato carboxilase (PEPcase), glutamina sintetase (GS), em nódulos, e a fixação biológica de nitrogênio, durante a ontogênese de feijoeiro (Phaseolus·vulgaris L.) e de caupi (Vigna unguicu1ata walp L.), foi rea1izado um experimento utilizando a simbiose Rhizobium legimunosarum bv phaseoli,/i>/Negro Argel e Bradyrhizobium sp./Vigna. As plantas foram cultivadas em casa de vegetação, em substrato sílica+vermiculita (1:1) e irrigadas com solução nutritiva isenta de N-mineral. Foram realizadas 10 amostragens destrutivas das plantas durante o ciclo de desenvolvimento das duas cultivares. Os parâmetros analisados em ambos simbiontes, foram: atividade das enzimas PEPcase, GS e nitrogenase nos nódulos; peso da matéria f:resca e número de nódulos; conteúdo de nitrogênio e de matéria seca nas raízes, caule+folha e vagens. De uma maneira gera1, as atividades das enzimas PEPcase e GS correlacionaram-se positivamente com a da nitrogenase e com a fixação simbiótica do nitrogênio (N2), através do peso da matéria fresca dos nódulos e do conteúdo de N total na planta. Em Negro Argel, as atividades das enzimas PEPcase, GS e nitrogenase apresentaram dois picos fases de florescimento e de enchimento médio das vagens durante toda a ontogênese, enquanto que, Caupi apresentou somente um pico na fase de enchimento das vagens. Os dados relatados neste experimento sugerem que a eficiência no processo de fixação de N2 depende, não somente da redução do N2 (enzima nitrogenase), mas também da recuperação do carbono evoluído (através de PEPcase) e da assimilação do nitrogênio fixado (através de GS).

Relationship between phosphoenolpyruvate carboxilase and glutamine synthetase activity and biological nitrogen fixation, during onthogenesis of nodulated bean (Phaseolus vulgaris L.) and cowpea (Vigna unguiculata walp L.) plants

The aim of this work to study the relationship between the activity of the enzymes phosphoenolpyruvate carboxilase (PEPcase) and glutamine synthetase (GS) with biological nitrogen fixation, in nodules, during the onthogenesis of bean (Phaseolus vulgaris L.) and Cowpea (Vigna unguiculata walp L.) plants. A greenhouse experiment was carried out using the symbiosis Rhizobium leguminosarum bv. phaseoli (CIAT-899)/Negro Argel and Bradyrhizobium sp. (CB-756)/Cowpea symbiosis. Plants were grown on a 1:1 silica/vermiculite·substrate and were watered with a nutrient solution without mineral-N. Ten samples of plants were take during the·growth cycle of both leguminous plants. The parameters analyzed in both cases were: activity of the enzymes PEPcase, GS and nitrogenase; number and fresh weight of nodules; nitrogen content and dry weight of roots, stalk+leaves and pods. In general, the activities of the enzymes PEPcase·and GS were positively correlated with that of nitrogenase and nitrogen fixation, as nodule fresh weigth and planto nitrogen content. In Negro Argel, the activities of PEPcase, GS and nitrogenase apresented two peaks during its entrie onthogenesis, one just before flowering and another during the mid pod filling stage. Cowpea, however, presented on1y one peak during "pre-flowering". The data reported in this experiment shows that the of N2 fixation process depends not only on also on the reduction of N2 (nitrogenase enzyme), but also on the assimilation of envolved carbon (PEPcase) and nitrogen which had been fixed (GS).
