O presente trabalho foi realizado com o objetivo de verificar a ação de agentes estressantes sobre o acúmulo e a degradação, no teor de trealose em Saccharomyces cerevisiae, utilizando a sacarose, sorbitol, cloreto de potássio e o etanol. Foram realizados quatro ensaios, sendo que em três deles utilizou-se leveduras de panificação encontradas no comércio, e no outro ensaio foram utilizadas as leveduras M-300-A e uma linhagem isolada da usina Jardest da safra de 1990. No primeiro ensaio foi realizado uma fermentação utilizando a levedura de panificação e diferentes concentrações (2, 4 e 8%) dos agentes estressantes a uma temperatura de 30°C. Foi observado um maior acúmulo no teor de trealose e menor teor de matéria seca no ensaio com KCL. Com os outros agentes estressantes utilizados (sacarose, sorbitol e etanol), não foram observadas diferenças significativas. No segundo ensaio a fermentação foi realizada com a levedura M-300-A, Jardest e com a de panificação a uma temperatura de 30°C. Os agentes estressantes foram adicionados em uma concentração que proporcionou uma pressão de 1 atm. Novamente o cloreto de potássio foi o agente estressante que mais influenciou o acúmulo de trealose, assim como o menor crescimento celular observado. Quanto as três linhagens de levedura utilizada, a M-300-A foi a que apresentou o maior acúmulo no teor de trealose e o maior crescimento celular. O terceiro ensaio foi realizado com o objetivo de verificar o efeito dos agentes estressantes na degradação endógena no teor de trealose em leveduras de panificação. Foi utilizado um meio composto por tampão citrato de potássio, 0,1M e pH 4,5 a 40°C durante um período de quatro horas. Previamente foi estimulado um acúmulo de trealose nas leveduras. Foram utilizadas as concentrações de 8, 16 e 24% de sorbitol. Não foi observada nenhuma alteração significativa na degradação endógena de trealose. O quarto ensaio foi realizado com o objetivo de verificar o acúmulo de trealose em leveduras de panificação pela ação da pressão osmótica exercida pelo sorbitol em concentrações de 8, 16 e 24%. O meio utilizado era composto por tampão citrato 0,1M e pH 4,5, contendo 5% de glucose. O ensaio foi conduzido a 40°C por um período de quatro horas. O acúmulo de trealose foi observado somente com o controle, atingindo até 17%. Nas concentrações de 8, 16 e 24% não foi observado um acúmulo significativo.

Effects of stressing agents on trehalose content in Saccharomyces cerevisiae

This research has been carried out aiming at assaying stressing agents action on the accumulation and on the endogenous degradation of trehalose in Saccharomyces cerevisiae of sorbitol, potassium chloride and ethanol stress. Four assays were done and including three different commercial backer's yeast M-300 A strain, and a isolate from Jardest sugar plant (1990 harvest season), one test. ln the first assay, fermentation was carried out by using baker's yeast in presence of concentrations (2, 4 and 8%) of the stressing agents at 30°C temperature. It has been observed high influence of KCI on trehalose accumulation together with low. No significant differences were observed when the stressing agents (sucrose, sorbitol and ethanol). ln the second assay, fermentation was carried out by using M-300-A, commercial baker's and Jardest at 30°C. The stressing agents concentration were those to give stressing agents at 1 atm. Again, KCl was the most affecting stressing agent on trehalose accumulation, and cell growth, for out three yeast strains tested. M-300 A had higher trehalose content and cell growth, compared to Jardest and commercial baker's yeast. The third assay was carried out to look at the effect endogenous trehalose degradation in trehalose accumulation estimulated baker's yeast, through stressing agents. The growth was buffered with potassium citrate buffer 0,1 M and pH 4,5. The fermentation was performed at 40°C for four hours. No significant change in endogenous trehalose degradation was observed, for all three different osmotical pressure tested. The fourth assay was carried out aiming at verifying trehalose accumulation in baker's yeast by sorbitol caused osmotic pressure (at 8, 16 and 24% concentrations) the used must was contained 5% of glucose and was buffered with citrate buffer 0,1 M, pH 4,5. The assay was kept at 40°C for four hours. Trehalose accumulation was observed only in the control treatment, reaching up to 17%. No significant accumulation was observed for the 8, 16 and 24% sorbitol treatments.
