O presente trabalho tem como objetivo estudar as relações entre os níveis de açúcares e atividade de invertases, verificando a influência da característica varietal sobre a relação sacarose-invertase em tecidos fonte-reservatório e as variações fisiológicas nas atividades dessas enzimas durante o desenvolvimento da cana-de-açucar. Para tanto foram selecionados quatro cultivares (CB41-76, NA56-79, SP70-1143 e SP71-799) representativos em termos de produção de sacarose. As amostragens foram realizadas aos 3, 4,5, 6 e 7 meses apos o plantio, no ciclo da cana-planta, determinando-se em cada época e em cada amostra de tecido (lâmina foliar, bainha e colmo) os níveis de açúcares solúveis totais, açucares redutores, sacarose, atividade de invertase ácida solúvel (pH 5,5), invertase neutra (pH7,0) e invertase ácida da parede celular. Comprovou-se que essas últimas formas isoenzimáticas de invertase, estão firmemente ligadas às paredes celulares dos tecidos de lâmina foliar, bainha e colmo. Elas consistem de enzimas diferentes em cada tecido, sendo distinguíveis com base no pH ótimo, respectivamente 3,8, 3,5 e 2,2. Em geral, os mais altos níveis de atividade de invertase foram encontrados nas lâminas foliares de todos os cultivares onde o teor de sacarose foi relativamente mais baixo. Inversamente, os mais baixos níveis de atividade foram detectados na bainha foliar onde se observaram altos teores de sacarose. No tecido do colmo a relação invertase-açúcar variou de acordo com a característica fisiológica, própria de cada cultivar. Dos quatro cultivares estudados a atividade de invertase foi maior no CB41-76 que apresentou o menor conteúdo de sacarose no colmo e os mais altos teores de açúcares redutores. Observou- se que as diferenças entre os cultivares CB41-76 e NA56-79 quanto aos teores de sacarose nas folhas e colmos estão localizados na bainha e não na lâmina foliar. Essa diferença foi atribuída à elevada eficiência de migração da sacarase das lâminas para a bainha e à baixa eficiência de migraçao desse açucar da bainha para os internódios no cultivar CB41-76. Uma condição inversa foi observada para plantas dos cultivares NA56-79, SP70-1143 e SP71-799. A elevada atividade de invertase ácida solúvel nos tecidos de lâmina foliar durante as fases iniciais do crescimento pode ser uma característica de um cultivar com baixo teor de sacarose no colmo. Por outro lado a presença da invertase neutra e invertase de parede celular em níveis elevados no final do ensaio parece determinar o tamanho do “pool” de hexoses e uma menor eficiência na migração desse açúcar. O cultivar SP71-799, comparado aos demais mostrou-se mais eficiente em termos de teores de sacarose apresentando teores baixos de açúcar redutor nos internódios. Os resultados sugerem que o perfil de atividade das enzimas invertases nos tecidos da folha, bainha e colmo poderão constituir-se em um dos mais importantes parâmetros bioquímicos para a seleção e melhoramento de genótipos de cana-de-açúcar. Comprovou-se ainda que as isoenzimas invertases exercem a função-chave na mobilização, utilização e acúmulo de sacarose na planta cana-de-açúcar.

Relations among sugar levels and invertase activity in tissue of sugar-cane cultivars

The present work has as objective the study of the relations between sugar levels and invertases activities, verifying the influence of varietal characteristics on the sucrose-invertase ratio in source-sink tissues and the physiological variations of the activities of these enzimes during sugar-cane development. For this, four cultivars (CB41-76, NA56-79, SP70-1143 e SP71-799) were selected, which are representative in terms of sucrose productivity. Samplings were made 3, 4, 5, 6 and 7 months after planting, during the cycle of plant cane, to perform in each sampling time and part of plant (leaf blade, sheath and stalks) measurements of the levels of total soluble sugars reducing sugars, sucrose, soluble acid invertase activities (pH 5.5), neutral invertase (pH 7.0) and cell wall invertases. It was shown that these last isoenzimatic forms of invertase are strongly bound to cell walls of leaf blade, sheaths and stem tissues. They consist of different enzymes in each tissue, being distinguishable by the optimum pH,3.8 , 3.5 and 2.2, respectivity. In general the high levels of invertase activity were found in leaf blades of all cultivars in which the sucrose level was relatively lower. On the contrary the lowest levels of activity were detected in the sheaths of high sucrose levels. In stalk tissue the invertase - sugar ratio varied according to the physiological characteristic of each cultivar. Among the four cultivars studied CB41-76 showed the highest invertase activity, the lowest sucrose content in the stalk and the highest reducing sugar content. It was observed that the differences between CB41-76 and NA56-79 with respect to sucrose content in leaves stalks are located at the sheaths. This difference was attributed to the high migration efficiency of sucrose from leaves to sheaths and the low efficiency of migration of this sugar from sheaths to internodes, in plants of NA56-79, SP70-1143 and SP71-799. The high acid invertase activity, soluble in the tissues of the leaf blades during the initial phase of growth, can be a characteristic of a cultivar with low sugar content in the stalk. On the other hand, the presence of neutral, cell wall invertase at high levels during the last sampling dates seems to define the size of the hexose pool and a lower efficiency of sucrose migration. Cultivar SP71-799, compared to all others, was more efficient with respect to sucrose contents because in presented low contents of reducing sugars at internodes. The results suggest that the enzime activity profile of invertase in leaf tissue, sheath and stalk could constitute one of the most important biochemical parameters for the selection and breeding of sugar cane genotypes. It wast also shown that the isoenzyme invertases exert a key function in the mobilization, utilization and storage of sucrose in the sugarcane plant.
