Com o objetivo de estudar a eficiência da fisação simbiótica do nitrogênio em feijoeiro (Phaseolus vulgaris) foram conduzidos quatro experimentos, nos anos de 1980 e 1981, no Centro de Energia Nuclear na Agricultura, Piracicaba, São Paulo. Foram utilizadas quatro cultivares de feijoeiro (Carioca, Aroana, Moruna, Rio Tibagi e Venezuela-350) e 5 estirpes de Rhizobium phaseoli (C-05; C-29 = 127 K 17; C-40 = CIAT 255; C-88 = SEMIA 487 e C-89 = CIAT 57). O primeiro experimento foi conduzido em câmara de crescimento em vasos de Leonard, enquanto que nos outros utilizou-se vasos de plásticos de capacidade de 2,5 𝓁, e o experimento ocorreu em casa-de-vegetação. Determinou-se os parâmetros de peso de matéria seca e teor de nitrogênio durante o crescimento das plantas. Como medida da eficiência da fixação do N2, estudou-se o gasto energético relacionado com a evolução do H2. Para isso, avaliou-se a atividade da nitrogenase (redução de C2H2) e evolução de H2 dos nódulos. Para a avaliação da redução do C2H2 foram utilizados dois sistemas: plantas inteiras intactas e raízes noduladas destacadas. A evolução do H2 foi determinada em nódulos destacados e em plantas inteiras intactas. Determinou-se então a eficiência relativa dos elétrons destinados ao complexo da nitrogenase (ER), isto é, os elétrons que são realmente utilizados na redução do N2 e não na evolução do H2. Detectou-se também quais estirpes expressaram a atividade da hidrogenase unidirecional independente de ATP em meio de cultura. Nos terceiro e quarto experimentos, realizou-se também a análise da evolução hidrogenase CO2 por raízes noduladas e nódulos destacados, a fim de se verificar a eficiência respiratória dos sistemas simbióticos em estudo. No quarto experimento, procurou-se verificar os efeitos da aplicação de pequenas doses iniciais de nitrogênio mineral (7 e 35 ppm de N como KN03) na eficiência da fixação do N2, visto que esse elemento sempre está presente no solo e pode inibir o processo de nodulação e fixação do nitrogênio. Nas condições desses experimentos pode-se concluir que: a. Observou-se o efeito da cultivar de P. vulgaris, da estirpe de R. phaseoli e da interação cultivar x estirpe no peso de matéria seca e teor de nitrogênio da parte aérea, raiz, nódulos, vagens e sementes. Houve também influência na atividade da nitrogenase (redução de C2H2), na evolução do H2 e na ER do complexo da nitrogenase. b. A presença da enzima hidrogenase na estirpe de bactéria não implicou necessariamente na maior eficiência delas na fixação do N2. A hidrogenase de R. phaseoli também não foi capaz de reciclar todo o H2 produzido e a máxima ER encontrada foi de 0,89. c. A ER dentro de cultivares, a ER dentro de estirpes e a ER dos sistemas simbióticos nos primeiros estágios do ciclo da planta sempre apresentou uma relação positiva com o teor de nitrogênio da planta. d. Não se encontrou relação entre: peso dos nódulos e fixação do nitrogênio; peso dos nódulos e redução do C2H2, observando-se grande variação da eficiência nodular entre cultivares e entre estirpes. e. Não houve relação entre o método de redução do C2 H2, analisado por cromatografia gasosa, e o teor de nitrogênio fixado, e isso foi atribuído ao desvio de elétrons utilizados na evolução do H2, que foi de 11% a 56%. f. A eficiência respiratória das raízes noduladas e dos nódulos destacados foi relacionada positivamente com o peso e teor de nitrogênio das plantas e com a ER do complexo da nitrogenase. g. O melhor desempenho das estirpes e das cultivares foi relacionado positivamente com a maior ER e eficiência respiratória. h. A aplicação de pequenas doses iniciais de nitrogênio mineral favoreceu o acumulo de matéria seca e de nitrogênio total, e não prejudicou a fixação do nitrogênio. Essas pequenas doses não afetaram a ER dos sistemas, mas aumentaram a eficiência respiratória das raízes noduladas. i. Existem sistemas simbióticos onde a principal limitação para o crescimento das plantas hospedeiras e a deficiência de nitrogênio, e em outros provavelmente é o suprimento de carbono. j. Os vasos de Leonard podem limitar o crescimento das plantas e prejudicar as pesquisas de seleção de Rhizobium em casa-de-vegetação.

Efficiency of symbiotic nitrogen fixation x H2 evolution x respiration by the bean nodules (Phaseolus vugaris L.)

There were conducted (at CENA, Piracicaba, São Paulo, Brazil), four experiments, in 1980 and 1981, to study the efficiency of symbiotic nitrogen fixation in beans (Phaseolus vulgaris L.). The bean cultivars utilized were Carioca, Aroana; Moruna, Rio Tibagi and Venezuela, 350; the Rhizobium phaseoli strains utilized were: C-05, C-29 = 127 K 17; C-40 = CIAT 57, C-88 = SEMIA 487, C-89 = CIAT 255). ln the first experiment the plants were grown in Leonard jars and were kept in a growth chamber. The other experiments were conducted under greenhouse conditions, and the plants were grown in plastic pots of 2,5 𝓁. The dry weight and total nitrogen content during the plant growth cycle were determined. The energetic cost of H2 evolution was used as a parameter on the nitrogen fixation efficiency study. With this purpose, it was considered the analysis of nitrogenase activity (C2 H2 reduction) and of H2 evolution by the nodules. There were utilized two systems to measure nitrogenase activity: intact plants and detached nodulated roots. The H2 evolution was measured on intact plant systems and with detached root nodules. From this, it was estimated the Relative Efficiency (RE) of electron transfer to N2 via nitrogenase. It was also detected if the Rhizobium strains presented the hidrogenase activity on a proper medium. ln the third and fourth experiments it was measured the respiratory CO2 efflux from freshly detached nodulated roots and from detached nodules, to calculate the respiratory efficiency of nitrogen fixation systems. In the fourth experiment, the plants were supplied with low quantities of mineral nitrogen (7 and 35 pprn N as KN03), in order to evaluate the effect of this element on the efficiency of nitrogen fixation, since the nitrogen is always present in soils and can inhibit plant nodulation and nitrogen fixation. Under the conditions of these experiments, it can be said that: a. There was an effect due to the Phaseolus cultivar, an effect due to the Rhizobium phaseoli strain and an effect due to the interaction cultivar X strain, on dry weight and total nitrogen content of aerial parts of plants, roots, nodules, pods and seeds. There was also an in fluence on nitrogenase activity (C2H2 reduction), on H2 evolution and on the RE of nitrogenase system. b. The detection of hydrogenase uptake, ATP independent, from the strain was not always related with the higher nitrogen plant content. The hydrogenase failed to recycle all the H2 produced via nitrogenase, and the maxim RE found was 0,89. c. The RE between cultivars, the RE between strains and the RE between symbiotic systems in the first stage of plant growth, were always positively related with the total plant nitrogen content. d. There was no relation between: dry weight of nodules and nitrogen fixation; dry weight of nodules and C2H2 reduction. There was great variability on nodule nitrogen fixation efficiency, influenced by both cultivars and strains. e. There was no relation between the results obtained by the method of C2 H2 reduction, analysed by gas chromatography, and the nitrogen plant content. This was probably due to the lost of electrons on the H2 evolution. The lost of electrons ranged from 11% to 56%. f. The respiratory efficiency of detached nodulated roots and detached nodules was positively related with the dry weight and nitrogen plant content, and was also positively related with RE of nitrogenase system. g. The good performance of the strains was positively related with RE and respiratory efficiency of the nodules; the good performance of the cultivars was positively related with RE and respiratory efficiency of nodules and nodulated roots. h. The supply with low quantities of mineral nitrogen in the beginning of plant growth cycle favored the increase of dry weight and plant nitrogen content, and it was not disadvantageous to nitrogen fixation. This low supply of nitrogen didnt affect the RE of the symbiotic systems, but increased the respiratory efficiency of nodulated roots. i. There are symbiotic systems where the main limitation to host plant growth in the beginning of plant growth cycle is the deficiency of nitrogen, while in other systems the main limitation to host plant growth probably is the supply of carbon. j. The conduction of experiments in Leonard jars may limit the plant growth and prejudice the research about Rhizobium selection under greenhouse conditions.
