Abstract in Portuguese
O objetivo deste trabalho foi analisar o comportamento bioquímico e citológico de células de cana-de-açúcar in vitro quando submetidas a diferentes doses de radiação gama. Células de cana-de-açúcar cv. NA 56-79 foram irradiadas com radiação gama 60Co, e cultivadas em meio de cultura condicionador de calos durante 45 dias. Findo o período determinou-se a massa de matéria fresca e a massa de matéria seca dos mesmos. Urna amostra de cada dose de irradiação (0,5 e 15Krads) foi separada para exame citológico (Microscopia Eletrônica), e o restante do material (calos) foi liofilizado para determinação das análises bioquímicas. A análise de crescimento mostrou que o mesmo foi inversamente proporcional à dose de radiação empregada após45 dias. Observou-se um declínio na massa de matéria fresca enquanto que a massa de matéria seca praticamente não variou. Com a irradiação de 5 Krads, as células; ao microscópio eletrônico apresentaram áreas de necrose e granulação de material citoplasmático. Quando o tratamento foi com 15 Krads destacaram-se mitocôndrias com formas aneladas, núcleos contorcidos e alongados distalmente e vacúolos com precipitação de material no seu interior. Análises bioquímicas mostraram que houve um pequeno aumento no teor de Nitrogênio Total nos calos irradiados com 5 Krads, enquanto que com 15 Krads houve um peque no decréscimo quando comparados com o controle. o teor de N - solúvel aumentou proporcionalmente à dose empregada e o teor de proteína apresentou um pequeno decréscimo em relação ao controle, não havendo, porém, grande variação desse último entre as dosagens de 5 e 15 Krads. O nível de aminoácidos permaneceu constante para os três tratamentos. Quanto aos carboidratos, os níveis de Açucares Solúveis Totais e Açúcares Redutores apresentaram. grande variação em relação ao controle. A análise de isoenzimas de esterases mostrou variações nos padrões de bandeamento nos três tratamentos, enquanto que os padrões de malato-desidrogenase permaneceram constantes para os dois primeiros tratamentos, variando apenas para o tratamento de 15 Krads.

Biochemical and cytological response of sugar cane (Saccharum spp.) cells to gamma radiation
Abstract in English
The objective of this research was to analyse the Biochemical and Cytological behavior of in vitro sugar cane cells under different doses of gamma irradiation. Cells of sugar cane cv. NA 56-79 were irradiated with 60Co ganuna radiation and cultivated . in callus conditioning culture media for 45 days. Fresh and dry mas ses were determined at the end of this period. A sample of each irradiation dose (0,5 and 15 Krad) was separated for cytological examination (Electron Microscope) and the balance of the material (callus) was liofilized for biochemical analysis. Growth analysis showed that it was inversely proportional to the radiation dose used after 45 days. A decrease in fresh matter was noted while the dry matter praE tically did not vary at 5 Krad irradiation. The cells sho wed necrotic areas and granulation of cytoplasmic material at the electron microscope, 15 Krad treatment showed ringed mitochondria, twisted nucleus and distally elongated vacuo les with precipitates in the interior. Biochemical analysis showed that there was a small increase in total nitrogen content of callus irradiated at 5 Krads, while at 15 Krad there was a small decrease when compared with the control. Soluble-N content increase in proportion to the dose used and the protein content showed a little decrease in relation to the control, however, there was no great variation between 5 and 15 Krad doses. The aminoacid level remained constant for the three treatmertts. As far as carbohydrates are concerned there was a large variation in relation to the control of Total-Soluble-Sugar and Reducing Sugar Levels. Analysis of isoenzymes from esterase showed variation in grouping patterns in the three treatments, while patterns for malato-dehydrogenase remained constant for the first two treatments. varying only in the 15 Krad treatment.
