Abstract in Portuguese
O objetivo do presente trabalho foi o de contribuir para o conhecimento da atividade da polifenol oxidase (PFO) e da peroxidase (PO) em algumas frutas e hortaliças bem como selecionar o melhor método pelo qual elas podem ser controladas. Com este propósito foi estudado o comportamento das atividades dessas enzimas, o efeito de alguns métodos de inativação (tratamento térmico, metabissulfito de potássio e ácido ascórbico) e a taxa de regeneração após processamento térmico (appertização), nas seguintes frutas e hortaliças em que o escurecimento enzímico constitue real problema no processamento: pera (var. Dágua), figo (var. Roxo de Valinhos), banana (var. Nanica), maçã (var. Golden Delicious), pêssego (var. Talismã), cenoura (var. Roxa), couve-flor (var. Bola de Neve), batata (var. Bintje) e palmito (Juçara ou Jiçara). A PFO foi considerada a enzima mais importante no escurecimento enzímico das frutas e a PO no das hortaliças. As atividades da PFO e PO das frutas e PFO das hortaliças apresentaram um crescimento linear durante a reação de escurecimento. Já nas hortaliças a PO apresentou atividade com crescimento exponencial no decorrer da mesma reação. Para a PFO das hortaliças e PO das frutas, o tratamento térmico foi considerado o melhor método de inativação. Tanto o ácido ascórbico como o tratamento térmico mostraram-se eficientes no controle da atividade da PO nas hortaliças. Já para a PFO das frutas, o metabissulfito de potássio foi mais eficiente, porém mostrou-se inadequado para controlar a atividade da peroxidase em quase todos os produtos estudados. Em termos gerais, nas frutas, a PFO apresentou maior resistência à inativação que a PO e no caso das hortaliças ocorreu o inverso, ou seja, a PO apresentou maior resistência que a PFO. O palmito destacou-se dos outros produtos estudados pela intensa atividade apresentada pela PFO e PO, o que explica a rapidez com que escurece durante o seu preparo para processamento. Observou-se ainda nesta hortaliça que tanto a atividade da PFO como a da PO são muito maiores na base do creme do que na ponta, o que sugere que estas duas partes deveriam ser processadas separadamente. Quanto a regeneração das enzimas após o processamento térmico (appertização), o fenômeno foi constatado somente no caso da PO que mostrou assim grande estabilidade às condições adversas encontradas durante aquele tratamento térmico. A PFO por sua vez, demonstrou ser uma enzima muito sensível, não se regenerando durante o tempo em que os produtos processados ficaram armazenados.

Study of enzymic activity of polyphenol oxidase and peroxidase in some fresh and canned fruits and vegetables

The objective of the present work was to contribute for the knowledge of the polyphenol oxidase (PPO) and peroxidase (PO) activities in some fruits and vegetable, as well to select the best method by which they can be controlled. With this purpose the behavior of these enzymes activities, the effect of some inactivation methods (heat treatment, potassium metabissulfite and ascorbic acid) and the regeneration rate after canning were studied in the following fruits and vegetables, in which the enzymic browning represents a great processing problem: pear (var. Dágua), fig (var. Roxo de Valinhos), banana (var. Nanica), apple (var. Golden Delicious), peach (var. Talismã), carrot (var. Roxa), cauliflower (var. Bola de Neve), potato (var. Bintje) and heart-of-palm (Juçara or Jiçara). The PPO was considered the most important browning enzyme of the fruits and the PO of the vegetables. The PPO and PO activities of the fruits and PPO of the vegetables presented a linear behavior during the development of the browning reactions. On the other hand, an exponential behavior was observed for the PO activity of the vegetables during the course of the same reactions. For the vegetables PPO and fruits PO, the heat treatment was considered the best method of inactivation. Both ascorbic acid and heat treatment showed to be effectives to control the PO activity in the vegetables. On the other hand, for the fruit PPO potassium metabissulfite was the most effective method, but did not control the PO activity in almost all products studied. In general, for the fruits, the PPO presented more resistance to be inactivated than peroxidase and in the case of the vegetables the inverse occurred, that is, PO was more resistant than PPO. Heart-of-palm differed from the others products studied due to the intense activity presented by PPO and PO enzymes, which explains the rapid darkening that occurs during its preparation for processing. It was also observed in this vegetable, that PPO and PO activities are much higher in the base than in the upper part of the product, suggesting that these two parts should be separately processed. Concerning to the enzyme regeneration after canning, the phenomenon was evident only in the case of the PO which showed great stability in adverse conditions found during heat processing. PPO, for his turn, showed to be a very heat sensitive enzyme, as no regeneration was observed during the storage time of the processed products.
