Os objetivos deste estudo foram: 1) propor uma metodologia de avaliação do meio de cultura para multiplicação de gemas de Eucalyptus spp; 2) descrever sintomas de deficiência de nutrientes minerais in vitro; e 3) diagnosticar o equilíbrio químico do meio de cultura, através do programa Geochem. (MATTIGOD & SPOSITO, 1979). Os híbridos utilizados foram: Eucalyptus grandis x E. tereticornis; E. Grandis x E. urophylla e E. urophylla x E. grandis. O meio de cultura líquido foi o JADS (Correia, 1993). O experimento foi com delineamento estatístico inteiramente casualizado, com 15 repetições para cada híbrido/tratamento. A duração do experimento foi de 21 dias, com avaliações a cada 7 dias. Os tratamentos foram: a) meio completo; b) sem N; c) sem P; d) sem K; e) sem Ca; f) sem Mg; g) sem S; h) sem Fe; i) sem B; j) sem Cu; k) sem Zn; l) sem Mn; m) sem Co; n) sem Mo. Foi suplementado em todos os tratamentos 0,2 mg/l de BA, 0,1 mg/l de ANA e 30 g/l de sacarose. O pH do meio foi de 58 ± 0,1, antes da autoclavagem. As Culturas foram mantidas temperatura de 26°C ± 2°C, fotoperíodo de 16 horas iluminância de 1000 lux. Pela análise dos resultados obtidos pode-se concluir que: A análise do balanço químico do meio de cultura e das concentrações dos nutrientes no material vegetal mostraram as diferentes exigências nutricionais para cada híbrido. Os híbridos se comportam de maneira diferenciada à deficiência de nutrientes comprovando que a absorção depende não somente da relação e/ou interação dos nutrientes mas também dos fatores genéticos. A metodologia apresentou respostas mais rápidas e típicas na diagnose e avaliação das deficiências quando comparadas com experimentos ex vitro. Muitos sintomas de deficiência in vitro foram novos em relação aos sintomas descritos nas condições ex vitro. A ausência de alguns elementos no meio de cultura não provocou sintomas de deficiência marcantes. Isto ocorreu em relação à maioria dos micronutrientes. Por outro lado, deficiências de N, Ca, S, Fe e Zn apresentaram sintomas semelhantes nos 3 híbridos. Através da comparação dos resultados das interações das concentrações dos nutrientes do material vegetal com as análises do programa Geochem, modelo teórico, foi possível constatar o acúmulo ou diminuição dos nutrientes na planta, diagnosticando-se a sua demanda e eficiência nutricional.

Diagnosis of mineral nutrient deficiency in three Eucalyptus spp. hybrids cultivated in vitro

The aims of this work were: 1) to propose a methodology to evaluate the culture medium for Eucalyptus spp. shoot multiplication; 2) to describe the symptoms of in vitro mineral nutrient deficiency; and 3) to diagnose the chemical equilibrium of the culture medium making use of the Geochem program (MATTIGOD & SPOSITO, 1979). The eucalyptus hybrids used in this work were originally obtained from experiments installed by CORREIA (1993): Eucalyptus grandis x E. tereticornis, E. grandis x E. urophylla e E. urophylla x E. grandis. The liquid culture medium utilized was based on JADS, described by CORREIA (1993). The statisticaI design used was completly randomized with 15 replicates for each hybrid/treatment. The evaluations were made every 7 days for a period of 21 days. The treatments were: a) complete; b) without N, c) without P, d) without K, e) without Ca, f) without Mg, g) without S, h) without Fe, i) without B, j) without Cu, k) without Zn, I) without Mn, m) without Co and n) without Mo. All treatments were supplements with: 0.2 mg/l BA; 0.1 mg/l NAA and 30 g/l sucrose. The pH was adjusted at 5.8 ± 0.1 before autoclaving. The cultures weremaintained at 26 ± 2°C, 16 hours photoperiod at iluminance of 1000 lux. The analysis of the results lead to the following conclusions: The analysis of the chemical equilibrium of the culture medium and of the mineral nutrient content of the vegetative material showed the different mineral nutrient demand for each hybrid. The hybrids behaved differently in relation to the mineral nutrient deficiencies showing that the absorption depends on both, the nutrients ratio and/or interactions, and the genetic background. In the diagnosis and evaluation of mineral deficiencies, this methodology showed faster and more typical results when compared to experiments ex vitro. Several deficiency symptoma observed in vitro were new in relation to those described under ex vitro conditions. The absence of some mineral nutrients did not induce marked visual symptoms. This ocurred for most micronutrients. However, the three hybrids showed similar symptoms for N, Ca, S, Fe and Zn deficiencies. The comparison of the results from the interactions among the mineral nutrient contents of the vegetative material and of the Geochem program analysis, theoretical model, allowed to observe the increase or the reduction of the nutrient content in the plant, indicating its demand and nutricional efficiency.
