O objetivo deste trabalho foi desenvolver metodologia adequada para coleta, extração, armazenamento e testes de viabilidade "in vitro" do pólen de Eucalyptus camaldulensis Dehn, Eucalyptus grandis Hill ex Maiden, Eucalyptus tereticornis Sm e Eucalyptus urophylla ST Blake. A coleta do pólen foi feita em flores em antese oriundas de ramos coletados com botões florais no início da abscisão do opérculo e mantidos em recipientes com água por 2 a 4 dias, em casa de vegetação. Dos métodos de extração estudados: a seco, em água, éter etílico, éter de petróleo e acetona, a extração a seco foi o método mais adequado. Foram utilizados os seguintes métodos de secagem: câmara seca (22°C constante) estufa (35°C e 40°C constantes) e a vácuo sob sílica-gel (temperatura ambiente). Verificou-se inteirações entre os mesmos e as espécies quanto a porcentagem de germinação do pólen. Apenas a secagem em estufa (40°C) mostrou-se prejudicial à maioria das espécies. A velocidade de secagem das estruturas estaminais variou entre as espécies, sendo maior para o E. urophylla e E. grandis e menor para o E. camaldulensis e E. tereticornis. Atribuiu-se esse comportamento a diferente natureza das espécies. O pólen seco a vácuo, em estufa (35°C) e sem secagem foi armazenado em "freezer" (-16°C) e em refrigerador o (4°C) em frascos de vidro (18 ml) hermeticamente fechados e colocados dentro de dessecadores. Após três meses de armazenamento verificou-se que a redução de umidade desempenhou papel importante na manutenção da viabilidade. A temperatura de -16°C foi mais adequada para o armazenamento quando acompanhada de redução de umidade do pólen. A viabilidade para todos os ensaios foi determinada através de testes de germinação "in vitro" empregando-se meio de cultura à base de ágar (0,8%) e sacarose (30%). Evidenciou-se também que a necessidade de quantidades adicionais de boro e cálcio depende de vários fatores dificultando a determinação de uma dose ideal. A temperatura de 25°C e umidade relativa de 99%, foram utilizadas na germinação do pólen, sendo 24 horas o período considerado corno o mais indicado para a avaliação de germinação. A viabilidade foi determinada através da contagem dos grãos germinados e não germinados em microscópio óptico com o aumento de 450 x em campos ao acaso sendo o procedimento mais adequado avaliar 300 grãos totais por repetição. A análise estatística foi feita através do x2 (qui-quadrado) devido a heterogeneidade das variâncias e a natureza binomial dos dados. A avaliação do comprimento do tubo polínico através do método de câmara clara mostrou não haver o efeito dos diferentes·tratamentos de secagem, todavia detectou-se variações existentes entre as espécies, sendo que o E. camaldulensis destacou-se com as maiores médias. Detectou-se diferenças entre a germinação para as espécies estudadas, onde o E. camaldulensis apresentou as maiores médias para grande parte dos ensaios.

Pollen handling and viability of Eucalyptus spp.

The objective of this work was to develop adequate methodology for Eucalyptus camaldulensis Dehn, Eucalyptus grandisa Hill ex Maiden, Eucalyptus tereticornis SM and Eucalyptus·urophylla. S.T. Blake pollen harvest, extraction, storage and "in vitro" viability tests. The pollen harvest was made from flowers at anthesis from branches collected with flower buds at the beggining of the operculum abscission and maintained in containers with water under greenhouse conditions for 2 to 4 days. Among the pollen extraction methods utilized, dry, water, ethyl and petroleum ether and acetone, the dry extraction was the most adequate. Drying before storage was made through: dry chamber (constant 22°C), kiln (constant 35°C and constant 40°C) and under vacuum with silica gel (at room temperature ± 25°C). It was observed that there were interaction among methods and species in relation to the pollen germination percentage, and only the drying in kiln (40°C) was harmfull to most of the species. The drying rate of the stamen varied among species, this was higher for E. urophylla and E. grandis and lower for E. camaldulensis and E. tereticornis. This was attributed to the nature of the species. Pollen without drying and with drying under vacuum with silica gel and in kiln (35°C) was stored in freezer (-16°C) and in refrigerator (4°C) in vials (18 ml) hermeticaly tight and kept ·under disiccator for 3 months. After storage it was observed that the reduction of the moisture of the stamen was very important for the viability maintenance. The storage at the freezer (-16°C) was the most adequate for the pollen storage when had the previous drying. The viability test was made through the pollen germination test "in vitro" utilizing the medium made up of 0,8% agar and 30% sucrose. The need to add boron calcium to the medium depend on several factors and this makes it difficult to determine the ideal amount of these elements to be added. The pollen germination was carried out under 25°C temperature and 99% relative moisture for 24 hours period. This period of 24 hours was determined to be the most indicated. The viability was determined through the counting of pollen grain germinated and non germinated with the use of a microscope with 450 x magnification in fields ramdonly distributed. This procedure was the most adequate to evaluate replicates of 300 pollen grains. The statistical analysis was made through the use of the x2(chi square) due to the heterogem of variances and the binomial nature of the data. The evaluation of the pollen tube lenght showed not have effects of the drying methods. However there were variations among species and E. camaldulensis pollen tube showed the larger lenghts. There were differences among species for the germination percentage and E. camaldulensis showed higher pollen germination rates.
