Com o objetivo de testar a influência do estado nutricional de plantas matrizes de Bauhinia forficata Link na calogênese in vitro, plantas obtidas de sementes germinadas em areia lavada, após queda das folhas cotiledonares, passaram a ser irrigadas com soluções nutritivas contendo diferentes níveis de cálcio, magnésio e potássio. As concentrações das soluções nutritivas forneceram os níveis completos de nutrientes e ¼X, ½X, 2X e 3X da solução completa de cada nutriente. A cada 15dias de tratamento, explantes dessas plantas (segmentos de caule sem gemas axilares) foram coletados, desinfestados e inoculados em meio de cultura sólido MS acrescido de 1,5mg BAP.L-1 e 30g sacarose.L-1. A calogênese em termos de massa de matéria fresca, seca e teor de proteínas foi afetada pelo estado nutricional em cálcio, magnésio e potássio das matrizes fornecedoras de explantes. A massa de matéria seca de calos foi maior em explantes de matrizes tratadas com baixos níveis de cálcio e principalmente com longa duração. Altos ou baixos teores de magnésio ou potássio afetaram negativamente a calogênese comparados com a solução nutritiva completa. O teor de proteína nos calos foi inversamente proporcional ao tratamento com cálcio e diretamente proporcional ao tratamento com potássio. Concentrações de magnésio superiores e inferiores aos da solução completa afetaram negativamente o teor de proteína dos calos.

Abstract in English
Aiming at testing the effect of nutritional status of Bauhinia forficata Link plants on calogenesis in vitro, plants obtained from sand germinated seeds, after fall of cotyledonal leaves, were irrigated with nutritive solutions containing different levels of calcium, magnesium and potassium. The nutritive solutions provided the complete leveI of nutrients and leveIs of 1/4X, 1/2X, 2X and 3X of the complete leveI solution. At every 15 days of treatment, explants of these plants (stem segments without axial buds) were collected, disinfected and inoculated on MS solid culture medium supplemented with 1.5mg BAP.L-1 and 30g sucrose.L-1. The calogenesis expressed as fresh and dry matter weight, and protein content was affected by the nutritional status on calcium, magnesium and potassium of the explant provider treated plants. The callus dry matter weight was higher for calli induced on expIants from low leveI calcium treated pIant and was treatment duration dependent. LeveIs of magnesium and potassium higher or lower than complete nutritive solution, atfected negatively the calogenesis. The protein content of calli was inversely proportionaI to calcium treatments and directly proportional to potassium treatments. LeveIs of magnesium higher and lower than complete solution affected negatively the protein content of the calli.
