O gênero Phaseolus, originado no Continente Americano compreende mais de 100 espécies e somente 4 delas (P. vulgaris, P. coccineus, P. acutifolium e P. lunatus) são economicamente cultiváveis. Entre os grãos, o feijão Phaseolus ocupa o 4º lugar em área cultivada e o 6º lugar em importância econômica. O Brasil ocupa o 1° lugar no mundo em produção bem como em consumo de feijão. Portanto, qualquer fator que afete a produção deste grão refletirá na esfera socioeconômica do País. O crescimento e a produção do feijão são afetados por doenças de origem bacteriana (Xanthomonas phaseoli e Pseudomonas phaseolicola), fúngica (Isariopsis griseola, Colletotrichum lindemuthianum, Macrophomina phaseoli, Uromyces phaseoli e Rhizoctonia sp.) e virótica (mosaico comum e mosaico dourado). Diferentes níveis de tolerância das espécies de Phaseolus cultiváveis aos agentes causais acima têm sido estudados. Este trabalho foi embasado na utilização do perfil eletroforético das isoenzimas para identificação de híbridos interespecíficos de Phaseolus, selecionados de cruzamentos de paternais que mostram tolerância ao vírus do mosaico dourado do feijoeiro. As isoenzimas de fosfatase ácida, peroxidase e esterase de 14 híbridos, obtidos de 6 cruzamentos diferentes envolvendo P. vulgaris e P. coccineus, foram separadas através de eletroforese em gel de poliacrilamida. Os resultados indicam que após a exposição das plantas ao vetor da doença no campo, a atividade das isoenzimas de peroxidase e fosfatase ácida foi sempre maior do que a observada antes da exposição. Resultado oposto foi observado para isoenzimas de esterase. Um perfil eletroforético de isoperoxidase é sugerido para ser usado como marcador bioquímico para tolerância ao BGMV, no feijão Phaseolus.

Perspectives on the utilization of isozymes for the identification of bean hybrids (Phaseolus sp.) resistant to bean golden mosaic virus

The genus Phaseolus, originated from American Continent, contains more than 100 especies and only four of them (P. vulgaris, P. coccineus, P. acutifolium e P. lunatus) at economically cultivated. Among Brazilian crops, Phaseolus beans are ranked 4° place in cultivated area and 6° place in cash revenue. Brazil is also ranked 1º in the world in common bean production and consumption. Therefore, any factor affecting this crop production will affect Brazilian economy to some extention. The growth and production of Phaseolus bean plants are affected by bacterial diseases (Xanthomonas phaseoli and Pseudomonas phaseolicola), fungal diseases (Isariopsis griseola, Macrophomina phaseoli, Uromyces phaseoli, Colletotrichum lindemuthianum and Rhizoctonia sp. and by virus diseases (common mosaic and golden mosaic). Different levels of tolerance to any the above disease agents by some of the cultivated especies of Phaseolus have been reported. This work was aimed at the utilization of isozyme electrophoretical to identify hybrids which show tolerance to the bean golden mosaic virus (BGMV). The isozymes peroxidase, acid phosphatase and esterase of 14 hybrids obtained 6 differents crosses involving P. vulgaris and P. coccineus were separated by polyacrilamide gel electrophoresis. The results indicated that after the hybrid plants were exposed to the disease vector in field, the activity of peroxidase and acid phosphatase isozymes was always higher in tolerant plants than in their susceptible counterpart. Opposite results were observed for esterase isozymes. An isoperoxidase electrophoretical profile that could be used as biochemical marker for BGMV tolerance in Phaseolus plants is suggested.
