Este trabalho teve por objetivo desenvolver um protocolo para a micropropagação do cravo, empregando a técnica de cultura de meristemas para limpeza clonal, contornando o problema da vitrificação. Para da tanto, determinaram-se as condições controladoras vitrificação do cravo in vitro, analisando o efeito de diferentes níveis do regulador BAP (benzilaminopurina), "Gelrite", ágar, nitrato de amônio, cloreto de cálcio e sistemas de vedação. Baixos níveis de BAP e nitrato de amônio aliados ao baixo potencial de água do meio, elevados níveis de cálcio e baixa umidade condicionada por tampas de algodão foram capazes de inibir a vitrificação. "Gelrite" revelou-se forte indutor do fenômeno em relação ao ágar. A taxa de vitrificação nos tratamentos de nitrato de amônio e cloreto de cálcio foi seguida por alterações na atividade da peroxidase. Em altos níveis de amônio, constatou-se baixa atividade da peroxidase, indicando o envolvimento da enzima no processo. Efeito contrário foi observado quando elevados níveis de cálcio estavam presentes. Melhores taxas de propagação foram promovidas por elevados níveis de BAP, alto potencial de água do meio e alta umidade, porém com elevado índice de vitrificação. Alterações na organização estrutural dos tecidos, como células hiperídricas, grandes espaços intercelulares e reduzido sistema vascular caracterizaram o material vitrificado. Foram testados os efeitos de ácido indoliacétio (AIA), ácido naftalenoacétio (ANA) e ácido indolbutírico (AIB) no enraizamento in vitro, onde todos os tratamentos promoveram a formação de raízes, mas não diferiram do controle, indicando a eliminação do uso de reguladores para esta fase. Com relação à aclimatação, plantas enraizadas in vitro foram confrontadas contra plantas não enraizadas e os resultados indicaram nenhum efeito no crescimento da parte aérea aos 90 dias em casa de vegetação, indicando a supressão da fase de enraizamento in vitro.

Biochemical and physiological parameters of the vitrification and development of carnation (Dianthus caryophyllus L.) in vitro
Abstract in English
The objective of this work vas the establishment of a protocol for micropropagation of carnation by using meristem culture technique to pursue clonal cleaning overcoming vitrification problems. It consisted on determining the carnation vitrification causes in vitro by assaying the effect of (BAP) benzyIaminopurine, "Gelrite", agar, ammonium nitrate, calcium chloride concentrations and of different covering system. Low levels of BAP and ammonium nitrate together with the low water potential, low moistness conditions and high level of calcium were able to inhibit vitrification. The vitrification rate in ammonium nitrate and calcium chloride treatments was monitored by the activity of peroxidase. At high level of ammonium when high vitrification occurred, a lower peroxidase activity was observed evidencing an association of this enzyme with the vitrification process. An opposite result was observed in medium containing high calcium concentration when low vitrification was observed. High level of BAP and high moistness content stimulated the micropropagation, even though a vitrification index was also high. Alterations in the tissue structure as hiperydrical cells, larger intercelular spaces and reduced vascular system were always observed in vitrificaI material. IAA (indoleacetic acid), NAA (naphtalene acetic acid) and IBA (indolebutyric acid) did not show any improvement of the in vitro root formation compared to the control, which showed root development. The acclimation of in vitro rooted plants was tested against non rooted plants. The results indicated the absence of previous rooting effect on the plants development after 90 days kept in greenhouse.
