Este trabalho teve por objetivo testar a influência do estado nutricional de matrizes de cravo (Dianthus caryophyllus L.) em nitrogênio, potássio, cálcio e magnésio no processo de calogênese in vitro. Mudas de cravo foram enraizadas em vasos contendo areia e irrigadas com água durante 15 dias. Tratamentos com 0X, ¼X, ½X, 1X e 2X da concentração dos nutrientes nitrogênio, potássio, cálcio e magnésio da solução nutritiva completa foram imediatamente iniciados. Ápices das plantas foram coletados aos 30, 45 e 60 dias após início dos tratamentos como fonte de explantes. Após a assepsia, explantes folha, entrenós e nó foram inoculados em meio de cultura sólido de Murashige & Skoog (1962), suplementado com 1,0 g de caseína hidrolizada, 2 μM de cinetina, 3 μM de 2,4-D e incubados em sala de crescimento sob condições de fotoperíodo de 16/8 horas e temperatura de 26°C ± 2. Sintomas de deficiência ou de excesso não foram aparentes nas plantas tratadas após 60 dias de tratamento. A calogênese avaliada em massa de matéria fresca e seca foi afetada pelo estado nutricional das plantas de cravo sendo dependente do explante e duração do tratamento. Tratamento com potássio e nitrogênio afetou a calogênese em explantes nó de maneira proporcional à concentração e duração dos tratamentos. Teores de cálcio e mangnésio inferiores e superiores àqueles da solução nutritiva completa favoreceram a calogênese em explantes entrenós e nó, a qual foi inversamente proporcional à duração do tratamento

Effect of nutritional status on N, K+, Ca2+ and Mg+2 carnation (Dianthus caryophyllus L., Caryophyllaceae) plants on calogenesis in vitro
Abstract in English
This work was aimed to test the influence of carnation plant (Dianthus caryophyllus L.) nutritional status on nitrogen, potassium, calcium and magnesium on calogenesis process in vitro. Carnation cuts were rooted in sand containing pots and moistuned with water for about 15 days. Treatments with 0X, ¼X, ½X, 1X e 2X the nitrogen, potassium, calcium and magnesium concentration of complete nutritive solutions were immediately started. Plant apexes were collected at 30, 45 and 60 days after treatment started as source of explants. After appropriate assepsy, leaf disc, internodes and node segments were inoculated on Murashige & Skoog (1962) solid culture media containing 1,0 g L-1 hidrolysed casein, 2 µM kinetin and 3 µM 2,4-D and incubated in growth room at 16/8 hours photoperiod and 26°C ± 2 temperature. No deficiecy and toxicity simptoms were apparent on the treated plants after then 60 days treatments. Calogenesis evaluated based on alli fresh and dry weigth was affected by carnation plants nutritional status for nutrient tested being explant and treatment duration dependent. Potassium and nitrogen treatment affected calogenesis induced on nodal explants in a concentration and treatment duration dependent fashion. Treatment levels of calcium and magnesium lowers or highers than those of nutritive complete solution favored calogenesis induction on internodes and node explant showing a treatment inversely proportionality pattern.
