A identificação de espécies de Trichogramma é feita principalmente através da análise da genitália do macho. Há, entretanto, a presença de espécies crípticas e/ou próximas, de difícil caracterização, além do fato do parasitóide ser muito pequeno (0,25 mm) e de difícil preparação em lâminas microscópicas. A técnica, apresentada neste trabalho, trato do sequenciamento da região ITS2 do rDNA com o objetivo de facilitar a identificação de espécies desse grupo de parasitódes. A técnica foi desenvolvida na Universidade de Wageningen, Holanda, e possibilita, em conjunto com dados taxonômicos, uma segura identificação das espécies utilizadas em programas de controle biológico aplicado. Utilizando-se essa técnica, é possível ainda a construção de uma chave molecular precisa, baseada em enzimas de restrição, juntamente com o comprimento de cada sequenciamento. Populações de diversas localidades do Brasil foram coletadas e mantidas em laboratório em condições controladas. A partir de populações de isofêmeas, cinco espécimens de cada população foram macerados em Chelex 5% e amplificados com “primers” específicos para a região ITS2 do rDNA. Cada DNA. Cada DNA foi ligado a um vetor pGEM®-T da Promega, sendo posteriormente plaqueados em meio LB para o crescimento de colônias brancas. A partir dessas colônias, foi obtido o “miniprep”, que foi enviado ao sequenciador. As sequências foram alinhadas com o auxílio do programa de computador ESEE 3.0. A partir dos resultados obtidos, uma chave molecular de algumas espécies brasileiras de Trichogramma. Comparando-se as seqüências obtidas com dados taxonômicos, confirmou –se a ocorrência no Brasil de T. lasallei Pinto, 1998 espécie recentemente registrada no País, baseado em características morfológicas. Utilizando-se um PCR com “primers” específicos, foi constatado pela primeira vez no Brasil, a presença de Wolbachia, em uma população telítoca de T. atopovirilia Oatman & Platner, 1983.

Identification of Trichogramma species (Hymenoptera: Trichogrammatidae) using the sequence of the ITS2 region of the rDNA
Abstract in English
Trichohramma identification nowadays is done basically by the analysis of the male genitalia. Nevertheless, there are cryptic or closely related species, which are difficult to identify using microscopic slides, and because they are very tiny (0.25 mm). The technique report in this work uses the sequence of the ITS2 region of the rDNA with the purpose to identify Trichogramma species. The technique was carried out at Wageningen University (Netherlands), and together with taxonomic methods, may allow a precise species identification used in biological control programs. By using this technique, it is possible to construct a precise molecular key based on restriction enzymes and on length of each sequence. Samples of Trichogramma populations from several regions of Brazil were collected and brought to the laboratory and reared under controlled conditions. From isofemale colonies, 5 individuals of each one were ground in Chelex 5% and amplified with specific primers for the ITS2 region of the rDNA. Each DNA was linked to a pGEM®-T vector by Promega and placed in Petri dishes with LB medium to grow white colonies. From these colonies a miniprep was obtained and sent to the sequencer. The sequences were aligned using the software ESEE 3.0. From the data obtained, a molecular key to some Brazilian species was elaborated. Comparing the sequences of each population and together with taxonomic data, it was confirmed the occurrence of T. lasallei Pinto, 1998 in Brazil, which had been recorded in this country recently, based on morphological characters. Also, this is the first record of a thelytokous population of T. atopovirilia Oatman & Palmer, 1983 infected with Wolbachia in Brazil, using PCR with specific primers.
