O objetivo da pesquisa foi verificar a adequabilidade de sementes de leguminosas e subprodutos agroindustriais, como componentes de meios para obtenção de esporos e cristais de Bacillus thuringiensis Berliner var.Kurstaki, com alta virulência e baixo custo. A partir da quantificação de esporos produzidos em 43 meios. foram selecionados 4 meios, incubados sob 31 ± 1°C, 120 rpm, em agitador-incubador, com acompanhamento de parâmetros de produção. Todos os meios produziram esporos, sendo que a maior produção (5,41 X 1010 esporos/ml) foi obtida no meio Soro de leite bovino + Leite de soja + Melaço de cana. O menor custo de produção foi obtido com o meio Farinha de crisálida + Melaço de cana. A atividade tóxica do complexo esporo-cristal foi afetada pela composição do meio, detectada por bioensaios com lagartas de Anticarsia gemmatalis Hubner(Lepidoptera, Noctuidae), sendo a menor Cl50 observada para o produto obtido no meio Caldo Nutriente + extrato de levedura.

Production of Bacillus thuringiensis Berliner var.Kurstaki on alternative media

This research was carried out to verify the feaeibility of leguminous seeds and agroindustrial by products as component of media used for the obtention of apores and crystals of Bacillus thuringiensis Berliner var.Kurstaki, with high virulence and low cost. From the quantification of apores produced on 43 media there were selected 4 media'which were incubated at 31 ± 1°C, 120 rpm, in a shaker, and registered the parameters of production. All these media have produced apores and the highest production (5,41 X 1010 spores/ml) was obtained on the medium cheese whey + soybean milk + sugarcane melasses. The lowest cost of production was obtained with the silk.worm pupa flour + molasses medium. The toxic activity of the spore-crystal complex was affected by the medium composition and detected through bioassays with Anticarsia gemmatalis Hubner(Lepidoptera, Noctuidae), larvae, being the lowest LC50 observed for the product obtained on the medium nutrient broth + yeaet extract.
