A possibilidade de utilização das reações sorológicas foi investigada com o objetivo de tornar mais prático e específico o diagnóstico de Bacillus thuringiensis em larvas de insetos, sejam estes espécies úteis, vetores de doenças ou pragas, suscetíveis àquele microrganismo. Um imunessoro foi obtido a partir de coelhos iimunizados com os antígenos flagelar, somático, capsular e do esporo de uma estirpe de B. thuringiensis var. kurstaki (soro tipo III) isolada de uma formulação comercial deste inseticida biológico e caracterizada previamente. No exame citobacterioscópico de culturas da referida estirpe, foi constatada a presença de formas vegetativas bacilares Gram-positivas, peritríquias e capsuladas. Através de um artifício utilizado na coloração de Gram, foram evidenciados esporos sub-terminais e isolados não corados, além de cristais dispersos, com estrutura bi-piramidal, semelhante à da ẟ - endotoxina do B. thuringiensis. Quanto ao aspecto cultural, a bactéria em questão proliferou em ágar simples com o aspecto de colônias rugosas, de bordo irregular e tendendo a formar um véu tipo pseudópodo. As colônias de B. thuringiensis apresentaram, em ágar sangue, um evidente halo de hemólise ao seu redor. A série bioquímica, convencionalmente empregada para a identificação de B. thuringiensis, foi complementada com uma prova de hidrólise da uréia, utilizando o meio de Uréia Stuart. O diagnóstico de B. thuringiensis através de reação de aglutinação rápida em lâmina e de reação de imunodifusão dupla radial em placa foi realizado em 155 larvas de insetos pertencentes à ordem Lepidóptera, constatando-se a ocorrência de bactérias morfologicamente pertencentes ao gênero Bacillus em 3.8% dos casos e a incidência de B. thuringiensis em 34.8% dos materiais examinados. A eficiência das reações sorológicas utilizadas foi comprovada pela determinação qualitativa da toxicidade das estirpes de B. thuringiensis isoladas. Os resultados obtidos permitiram concluir que: 1 - há possibilidade de empregar reações sorológicas específicas no diagnóstico de B. thuringiensis em larvas de insetos; 2 - a reação sorológica de aglutinação rápida em lâmina é a conselhável apenas para um diagnóstico rápido de triagem B. thuringiensis; 3 - a reação sorológica de imunodifusão dupla radial em placa fornece resultados de diagnóstico do B. thuringiensis, mais rápidos, mais específicos e de menor custo em relação à identificação bioquímica; 4 - as reações sorológicas para diagnóstico do B. thuringiensis devem ser executadas com um imunessoro obtido contra um ou mais antígenos de maior especificidade que o antígeno flagelar.

The possibility of utilizing serological reactions was investigated in order to make Bacillus thuringiensis diagnosis in insect larvae including beneficial species, vectors of maladies and pests more practical and specific. For the preparation of antiserum, rabbits were immunized with flagellar, somatic, capsule and spore antigens from a previously characterized strain of B. thuringiensis var. kurstaki (serotype III), isolated from a commercial formulation. The presence of Gram-positive, peritrichously flagellated and capsuled vegetative rods was observed through the citobacterioscopic test of a reference strains culture. Unstaine, subterminal and isolated spores, besides dispersed crystals of bipyramidal structure similar to B. thuringiensis ẟ - endotoxin were detected by means of a device used in Gramss stain. Regarding the cultural aspects, the microrganisms originated swarming and rough colonies on nutriente agar. Colonies upon blood agar exhibited a distinct zone of hemolysis. The biochemical series employed conventionally for identifying B. thuringiensis were complemented with a reaction of urea hydrolysis employing Stuarts urea medium. B. thuringiensis diagnosis through a slide agglutination test and radial double immunodiffusion in plate was carried out in 155 Lepidoptera insect larvae. It could be noticed that there was an occurrence of bacteria morphologically belonging to the genus Bacillus in 3.8% of the cases and the incidence of B. thuringiensis in 34.8% of the examined materials. The efficiency of serological reactions was proved by determining the qualitative toxicity of B. thuringiensis isolates. The results obtained lead us to conclude that: 1 - it is possible to use specific serological reactions for the diagnosis of B. thuringiensis in insect larvae; 2 - the slide agglutination test is suitable only for a B. thuringiensis rapid screening diagnosis; 3 the radial double immunodiffusion in plate gives faster, more specific and cheaper results of B. thuringiensis diagnosis, in relation to the biochemical identification; 4 - the serological reactions for B. thuringiensis diagnosis must be done with an antiserum against one or more antigens, more specific than the flagellar antigen.
