O Alphavirus Mayaro (MAYV) é um arbovírus conhecido como causador de doença febril aguda humana, que vem emergindo no Brasil com potencial para ser causador de grave problema de saúde pública. Sabendo que alguns Alphavirus são causadores de doença grave no recém-nascido por transmissão vertical, decidimos estudar o vírus Mayaro. Avaliamos a infecção transplacentária causada por MAYV em camundongos fêmeas C57/BL6j. Assim, analisamos a infecção por MAYV em diversos períodos de gestação dos camundongos fêmeas e suas possíveis implicações. Buscamos identificar a infecção por MAYV em placentas, cérebro e outros órgãos de mães infectadas e seus fetos. Para tanto, foram analisados 8 grupos de camundongos fêmeas prenhes que foram infectadas com altos teores de MAYV (107 UFP/100µL) em diferentes fases gestacionais. Placentas, cérebros e macerados de órgãos fetais e órgãos maternos que foram analisados por RTq-PCR amplificando uma região do gene 5' UTR e NSP1 do MAYV. Foi também, analisado a infecção perinatal e as manifestações clínicas em animais recém-nascidos de mães infectadas por MAYV. Os resultados mostraram que o modelo experimental de infecção congênita por vírus Mayaro em camundongos é viável e pode ser reproduzido em estudos com outros vírus. O vírus Mayaro é transmitido por via transplacentária em camundongos C57BL/6j causando infecções congênitas graves. O vírus Mayaro infecta e por via transplacentária acomete os fetos murinos em diferentes fases da gestação.

The Alphavirus Mayaro (MAYV) is an arbovirus known to cause acute febrile human disease, which has been emerging in Brazil with the potential to cause a serious public health problem. Knowing that some Alphaviruses are known to cause severe disease in newborns by vertical transmission, we decided to study the Mayaro virus. We evaluated the transplacental infection caused by MAYV in C57/BL6j mice. Thus, we analyzed the MAYV infection in different periods of gestation of the mice and its possible implications. We sought to identify MAYV infection in placentas, brains, and other organs of infected mothers and their fetuses. To this end, 8 groups of pregnant mice were infected with high titers of MAYV (107 PFU/100µL) at different gestational stages. Placentas, brains, and macerates of fetal and maternal organs were collected and analyzed by RTq-PCR amplifying a region of the 5' UTR and NSP1 genes of MAYV. The perinatal infection and clinical manifestations in newborn animals from MAYV-infected mothers were also analyzed. The results showed that the experimental model of congenital infection by Mayaro virus in mice is viable and can be reproduced in studies with other viruses. The Mayaro virus is transmitted transplacentally in C57BL/6j mice causing severe congenital infections. The Mayaro virus infects and transplacentally affects murine fetuses at different stages of gestation.
