The aim of this in vitro study was to evaluate the micro-shear bond strength and the roughness to human dentin using different instrumentations (SiC #600, diamond rotary bur, CVDt and CVDi) and three dentin adhesive/composite resin systems: Adper Sigle Bond 2 (SB), Clearfil SE Bond (CF) e Adper Prompt L-Pop (PL). Seventy-two third molars were split in the middle. The oclusal third from the one hundred and forty and four halves were removed and standardized using the SiC #600. After the prepared surfaces, which were analyzed the roughness average (Ra) with a surface perfilometer and hybridization. The specimens were then stored in distillated water for 24 hours at 370C. The specimens were then were attached to a micro-shear apparatus and subjected to a micro-shear strength test at a crosshead speed of 1mm/min until failure occurred. The two-way ANOVA indicated that there was significant differences in average roughness between the instrumentations which SiC #600 was the smoothest surface. No differences were detected between dentin surface prepared with diamond bur and CVDi (intermediate in the roughness assessment) and the CVDt gave the roughest surface. The bond strengths varied slightly, though dentin instrumentations with CVDi showed the lowest values than the surfaces prepared with SiC #600 and diamond bur. The bond strengths varied slightly, though adhesives, PL showed lower micro-shear bond strength than SB and CF, which were no significant relevance. The interaction of SiC #600, diamond bur and CVDt using SB and CF showed no significant difference in micro-shear bond strength and higher than PL. The instrumentation showed no significant differences in micro-shear bond strength using CF and PL.
