Treatments for dentin hypersensitivity (DH) do not exhibit long-term effectiveness, what boost the search for new treatments. The use of bioglass that is a biostimulator material with the ability to promote hydroxyapatite deposition would be a promising treatment for long term DH reduction. In this sense, the aim of this study was to compare the in vitro effect of a bioglass powder in different vehicles for closing exposed dentinal tubules before and after an erosion / abrasion challenge. Two fragments of the cervical portion of the root were obtained from 36 single-rooted human teeth. The cementum was removed with periodontal curettes. Then, to mimic the DH, the dentin was treated with 37% phosphoric acid for 1 min to open the tubules. The fragments were assigned to two experimental groups (n = 12): G1: Control: no additional treatment; G2: bioglass with 30% phosphoric acid, G3: Gluma Desensitizer; G4: Gluma Desensitizer with bioglass; G5: Nd:YAG laser irradiation and G6: bioglass with Nd:YAG laser irradiation. Irradiation was performed with Nd: YAG laser (1.064 nm, 1.0 W, 15 Hz, 66 mJ, 10s, four times with 10s- intervals between them) in scanning and contact mode. After the treatments, all specimens from different experimental groups were stored into artificial saliva for 14 days. The two fragments of the same roots received the same treatment and at random, one fragment was submitted to erosion/abrasion challenge and the other not. This challenge was carried out by the immersion of the specimens into a 1% citric acid solution (pH 3.75) for 2 min, followed by immersion into 15 ml of artificial saliva for 60 min. Next, the specimens were submitted to brushing (270 cycles in slurry of toothpaste and artificial saliva for each specimen) and washed in the end of this challenge. These procedures were repeated three times a day for 3 days. Then, the specimens were stored in artificial saliva for 24 h. After incubations in artificial saliva all specimens of different experimental groups were prepared for morphological analysis by scanning electron microscopy (SEM). The scanning electron micrographs were analyzed qualitatively and quantitatively, and the relative area of open tubules in relation to the total area of the images with and without the erosion/abrasion challenge were compared by using the Student´s t test (p < 0.05). The major morphological changes of the dentine were observed in the Nd:YAG groups and in the bioglass + 30% phosphoric acid group. The Nd:YAG groups were not considered for qualitative analysis because irradiation was uneven. The Gluma group without erosion/abrasion challenge showed relative area of open tubules significantly lower than those of all other groups with and without erosion/abrasion challenge. We conclude that the bioglass powder in different vehicles did not induce obliteration of exposed dentinal tubules.
